尼可地爾抑制野百合堿誘導(dǎo)的肺動(dòng)脈高壓小鼠肺血管重構(gòu)

【摘要】:目的 :研究ATP敏感性鉀離子通道開(kāi)放劑尼可地爾對(duì)野百合堿誘導(dǎo)的肺動(dòng)脈高壓小鼠肺血管重構(gòu)的影響。方法 :CD1雄性小鼠隨機(jī)分為對(duì)照組、野百合堿模型組、野百合堿+尼可地爾治療組,每組6只,后2組給予野百合堿(400 mg/kg)皮下注射,每周1次,連續(xù)8周,制備肺動(dòng)脈高壓模型,治療組每日給予尼可地爾(10.5 mg/kg)灌胃。8周后通過(guò)直接經(jīng)膈肌右心室穿刺法測(cè)定右心室收縮壓;稱(chēng)重法計(jì)算右心肥厚指數(shù);HE染色觀察右室心肌細(xì)胞形態(tài);彈力纖維染色測(cè)定肺小動(dòng)脈中膜厚度;免疫組化檢測(cè)肺動(dòng)脈平滑肌細(xì)胞α-肌動(dòng)蛋白(α-smooth muscle actin,α-SMA)表達(dá)并計(jì)算肺小動(dòng)脈肌化程度。結(jié)果:尼可地爾(10.5 mg/kg)能顯著降低野百合堿誘導(dǎo)的小鼠右心室收縮壓升高(P0.01)和右心肥厚(P0.05),減輕肺小動(dòng)脈中膜增厚(P0.01),抑制肺小動(dòng)脈完全肌化(P0.01),減輕右心心肌細(xì)胞損傷。結(jié)論 :尼可地爾能通過(guò)抑制肺血管重構(gòu)減輕野百合堿誘導(dǎo)的肺動(dòng)脈高壓,其可能是一種能有效防治肺動(dòng)脈高壓的藥物。
[Abstract]:Objective: To study the effect of ATP sensitive potassium channel opener nicorandil on pulmonary vascular remodeling in pulmonary hypertension induced by monocrotaline. Methods: CD1 male mice were randomly divided into control group, monocrotaline model group, monocrotaline + nicorandil treatment group, 6 rats in each group, and 2 groups were given subcutaneous injection of monocrotaline (400 mg/kg), each group was given subcutaneous injection. The pulmonary hypertension model was prepared for 8 weeks on week 1, and the treatment group was given the right ventricular systolic pressure by direct phrenic right ventricle puncture after.8 weeks. The right ventricular hypertrophy index was calculated by weighing method, the right ventricular myocyte shape was observed by HE staining, and the thickness of the middle pulmonary artery in the pulmonary artery was measured by HE staining. The expression of alpha actin (alpha -smooth muscle actin, alpha -SMA) in pulmonary artery smooth muscle cells was detected by histochemistry and the degree of pulmonary arteriolating was calculated. Results: nicorandil (10.5 mg/kg) could significantly reduce the right ventricular systolic pressure (P0.01) and right heart thickness (P0.05) induced by nilrotaline, reduce the thickening of the pulmonary arterioles (P0.01) and reduce the lung small size of the pulmonary arterioles. Conclusion: Nicorandil can reduce pulmonary arterial hypertension induced by monocrotaline by inhibiting pulmonary vascular remodeling, and it may be an effective drug to prevent pulmonary hypertension.
