硫化氫對低氧性肺動脈高壓大鼠P47phox表達的影響

【摘要】:目的 觀察硫化氫H2S對低氧性肺動脈高壓(HPH)大鼠P47phox表達的影響。方法 36只體重(180±20)g健康成年雄性SD大鼠,隨機分為常氧組、低氧組、低氧+硫氫化鈉(Na HS)組,每組12只。低氧16 d后分別檢測其平均肺動脈壓(m PAP)、右心室肥厚情況和肺血管形態(tài)學(xué)指標(biāo)的變化。采用Western印跡檢測肺動脈組織P47phox的表達水平。結(jié)果 與常氧組比較,低氧組m PAP明顯升高,右室/(左室+室間隔)比值顯著增加(P0.01);部分肌型動脈百分比明顯升高,非肌型動脈百分比明顯降低(P0.05);肺動脈組織P47phox表達升高(P0.05)。與低氧組比較,低氧+Na HS組m PAP、右室/(左室+室間隔)比值均明顯降低(P0.01);肺小血管中肌型動脈、部分肌型動脈百分比明顯降低,非肌型動脈百分比明顯升高(P0.05);肺動脈組織P47phox表達明顯降低(P0.01)。結(jié)論 H2S可能通過抑制大鼠肺動脈組織P47phox表達而調(diào)節(jié)低氧性肺血管結(jié)構(gòu)重建和HPH的形成。
[Abstract]:Objective to observe the effect of hydrogen sulfide (H 2S) on the expression of P47phox in hypoxic pulmonary hypertension (HPH) rats. Methods Thirty-six healthy male SD rats with body weight of 180 鹵20 g were randomly divided into two groups: normoxic group (n = 12) and hypoxic sodium sulphide group (n = 12). After hypoxia for 16 days, the mean pulmonary artery pressure (MPP), right ventricular hypertrophy (RV) and pulmonary vascular morphology were measured. The expression of P47phox in pulmonary artery was detected by Western blot. Results compared with the normoxic group, m PAP was significantly increased in hypoxia group, the ratio of right ventricular septum (left ventricular septum) was increased significantly, the percentage of partial muscular artery was increased significantly, the percentage of non-muscular artery was significantly decreased, and the expression of P47phox in pulmonary artery was increased (P 0.05). Compared with the hypoxic group, the ratios of m PAPand right ventricular septum (left ventricular septum) in hypoxic Na HS group were significantly lower than those in hypoxic group, and the percentage of muscular artery and partial muscular artery in small pulmonary vessels were significantly decreased. The percentage of non-muscular arteries increased significantly, and the expression of P47phox in pulmonary artery decreased significantly (P 0.01). Conclusion H2S may regulate hypoxic pulmonary vascular remodeling and HPH formation by inhibiting the expression of P47phox in rat pulmonary artery.
