基于毛細(xì)管電泳的地中海貧血分析系統(tǒng)研制

【摘要】:地中海貧血是一種高發(fā)于我國長江以南地區(qū)的遺傳性溶血性疾病,通過檢測血液中血紅蛋白變異體的存在而確定地中海貧血病的高效毛細(xì)管電泳法是目前醫(yī)用最常規(guī)的地貧篩查手段。但是目前國內(nèi)市面上的醫(yī)用毛細(xì)管電泳儀多為外國生產(chǎn),而且體積龐大,價格昂貴。因此,本課題自主研發(fā)制造了一套基于毛細(xì)管電泳的地中海貧血分析系統(tǒng),以填補(bǔ)國內(nèi)毛細(xì)管電泳儀器開發(fā)領(lǐng)域的空白。首先,對高效毛細(xì)管電泳法進(jìn)行原理性研究,并結(jié)合地中海貧血檢測的相關(guān)特征,提出毛細(xì)管電泳系統(tǒng)的設(shè)計需求和設(shè)計方案。然后,根據(jù)系統(tǒng)設(shè)計方案完成系統(tǒng)機(jī)械結(jié)構(gòu)、系統(tǒng)液路結(jié)構(gòu)、系統(tǒng)光路結(jié)構(gòu)、系統(tǒng)硬件電路結(jié)構(gòu)和系統(tǒng)軟件的設(shè)計。其中,系統(tǒng)機(jī)械結(jié)構(gòu)和系統(tǒng)液路的設(shè)計滿足毛細(xì)管電泳系統(tǒng)框架與密閉性的需求。系統(tǒng)光路設(shè)計滿足地中海貧血樣品中血紅蛋白特異性波長檢測的需求。系統(tǒng)硬件電路中:可編程供電電源電路滿足系統(tǒng)各模塊供電需求;可編程高壓電源電路滿足系統(tǒng)電泳電壓0-12 kV連續(xù)可調(diào)需求;精密溫控電路滿足控制系統(tǒng)電泳溫度37℃需求;運動控制電路滿足系統(tǒng)自動化和毛細(xì)管nL級別進(jìn)樣需求;檢測電路滿足系統(tǒng)極微弱光檢測需求。最后,使用KeilμVision5編寫下位機(jī)驅(qū)動軟件,使用Visual Studio 2015編寫上位機(jī)界面軟件,并對系統(tǒng)軟硬件電路進(jìn)行上下位機(jī)聯(lián)合調(diào)試。在系統(tǒng)平臺搭建完成后,進(jìn)行系統(tǒng)關(guān)鍵模塊,包括高壓電源模塊、溫度控制模塊和檢測模塊的測試。在測試合格后進(jìn)行系統(tǒng)精密度和靈敏度的測試,并通過重復(fù)多次質(zhì)控樣品實驗對系統(tǒng)電泳電壓、進(jìn)樣量、進(jìn)樣濃度與光源強(qiáng)度等多個參數(shù)進(jìn)行優(yōu)化。在系統(tǒng)優(yōu)化完成后進(jìn)行地中海貧血血樣上機(jī)實驗,包括質(zhì)控液毛細(xì)管電泳實驗、α-地中海貧血毛細(xì)管電泳實驗和β-地中海貧血毛細(xì)管電泳實驗。實驗結(jié)果表明,本課題設(shè)計的毛細(xì)管電泳系統(tǒng)在電泳電壓9800 V、樣品稀釋6倍時靈敏度最高;系統(tǒng)精密度小于2%,滿足地中海貧血血紅蛋白檢測需求;且本系統(tǒng)能夠成功完成典型的α-地貧和β-地貧血樣分析檢查。
[Abstract]:Thalassemia is a highly prevalent hereditary hemolytic disease in the area south of the Yangtze River in China. High performance capillary electrophoresis (HPCE), which detects the presence of hemoglobin variants in blood, is the most common screening method for thalassemia. But at present, most of the medical capillary electrophoresis instruments on the domestic market are produced by foreign countries, and the volume is huge and the price is expensive. Therefore, a new analytical system of thalassemia based on capillary electrophoresis was developed to fill the blank in the field of capillary electrophoresis in China. Firstly, the principle of high performance capillary electrophoresis (HPCE) was studied, and the design requirements and scheme of capillary electrophoresis system were put forward according to the characteristics of thalassemia detection. Then, according to the system design scheme, the system mechanical structure, the system fluid structure, the system optical circuit structure, the system hardware circuit structure and the system software design are completed. Among them, the mechanical structure and liquid circuit design of the system meet the requirements of capillary electrophoresis system framework and sealing. The systematic optical circuit design meets the requirement of hemoglobin specific wavelength detection in thalassemia samples. In the hardware circuit of the system, the programmable power supply circuit can meet the demand of each module of the system, the programmable high voltage power supply circuit can meet the requirements of the system electrophoretic voltage 0-12 kV continuously adjustable, the precision temperature control circuit meets the requirement of the control system electrophoresis temperature 37 鈩,
