微小RNA-152介導(dǎo)葉酸對(duì)體外血管平滑肌細(xì)胞增殖遷移的抑制作用

【摘要】:目的探究葉酸對(duì)(folic acid,FA)血小板源生長(zhǎng)因子(PDGF-BB)誘導(dǎo)的血管平滑肌細(xì)胞(VSMCs)增殖遷移的影響及其機(jī)制。方法體外用PDGF-BB誘導(dǎo)大鼠血管平滑肌細(xì)胞系A(chǔ)7r5增殖遷移,分為對(duì)照組、PDGF組、PDGF+FA組、PDGF+FA+miRNA-152 inhibitor組、PDGF+FA+miRNA-152 NC組。采用實(shí)時(shí)定量聚合酶鏈反應(yīng)(RT-PCR)檢測(cè)各組VSMCs內(nèi)miR-152的表達(dá)水平。采用CCK-8試劑盒檢測(cè)各組VSMCs增殖水平。采用劃痕實(shí)驗(yàn)檢測(cè)各組VSMCs遷移水平。采用蛋白印跡法檢測(cè)細(xì)胞增殖核抗原(PCNA)蛋白、基質(zhì)金屬蛋白酶9(MMP-9)的表達(dá)水平。結(jié)果 RT-PCR結(jié)果顯示PDGF干預(yù)降低VSMCs miR-152表達(dá)水平,而PDGF+FA組VSMCs miR-152表達(dá)水平顯著高于PDGF組(P0.05)。PDGF干預(yù)VSMCs的增殖遷移能力顯著增強(qiáng)相對(duì)于對(duì)照組(P0.05),而PDGF+FA組的VSMCs增殖遷移能力低于PDGF組(P0.05),PDGF+FA+miR-152 inhibitor組的VSMCs增殖遷移能力強(qiáng)于PDGF+FA組(P0.05)。結(jié)論 miR-152參與介導(dǎo)FA抑制PDGF-BB誘導(dǎo)的VSMCs增殖遷移作用。[營(yíng)養(yǎng)學(xué)報(bào),2017,39(5):484-488]
[Abstract]:Objective to investigate the effect of folic acid on proliferation and migration of vascular smooth muscle cells (VSMCs) induced by platelet derived growth factor (PDGF-BBB) and its mechanism. Methods the proliferation and migration of rat vascular smooth muscle cell line A7r5 were induced by PDGF-BB in vitro, and were divided into two groups: control group (control group), PDGF-FA miRNA-152 inhibitor group (PDGF-FA miRNA-152 inhibitor group) and PDGF-FA miRNA-152 group (NC group). Real-time quantitative polymerase chain reaction (RT-PCR) was used to detect the expression of miR-152 in VSMCs. CCK-8 kit was used to detect the proliferation of VSMCs in each group. The migration level of VSMCs in each group was measured by scratch test. The expression of matrix metalloproteinase 9 (MMP-9) and proliferating nuclear antigen (PCNA A) were detected by Western blot. Results the results of RT-PCR showed that PDGF intervention decreased the expression of VSMCs miR-152. However, the expression of VSMCs miR-152 in PDGF FA group was significantly higher than that in PDGF group (P 0.05). The proliferation and migration ability of VSMCs in PDGF FA group was significantly higher than that in control group (P 0.05), while the VSMCs proliferation and migration ability in PDGF FA group was lower than that in PDGF group (P 0.05) and that in PDGF FA group was higher than that in PDGF FA group (P 0.05). Conclusion miR-152 is involved in the inhibition of VSMCs proliferation and migration induced by PDGF-BB mediated by FA. [Journal of Nutrition: 484-488]
