不同血氧飽和度影響主肺動脈壓力變化的離體血管模型研究

【摘要】:目的:建立不同血氧飽和度影響主肺動脈壓力變化的離體模型,為進一步探索肺動脈高壓機制提供研究平臺。方法:取新西蘭大耳白兔20只,隨機選取5只作為正常對照組。其余15只分離肺動脈后將血管置于培養(yǎng)液中,并接入體外循環(huán)系統(tǒng);以新鮮肝素化兔血灌流培養(yǎng)48 h,灌注壓力維持40 mm Hg(1 mm Hg=0.133 k Pa);期間通過調(diào)節(jié)膜肺給氧量控制血氧飽和度,建立3個模型組(n=5):高飽和度組(90%~100%)、中飽和度組(65%~75%)、低飽和度組(40%~50%)。轉(zhuǎn)流結(jié)束后獲取離體血管和正常對照組兔肺血管行病理形態(tài)分析、肺動脈組織IPP6.0定量分析以及勻漿一氧化氮含量分析比較。結(jié)果:3個模型組轉(zhuǎn)機時間均達到48 h,壓力穩(wěn)定控制在40 mm Hg,血氧飽和度由高到低分別為(97.94±1.01)%、(72.14±12.85)%、(43.83±8.71)%(P0.05)。病理分析結(jié)果顯示:與正常對照組兔肺動脈相比,3個模型組離體培養(yǎng)的肺動脈管壁均明顯增厚;隨著血氧飽和度降低,3個模型組離體培養(yǎng)的肺動脈彈力纖維逐漸增多、增粗,差異均有統(tǒng)計學意義(P0.05)。結(jié)論:本實驗建立了不同血氧飽和度影響主肺動脈壓力變化的離體血管模型。
[Abstract]:Aim: to establish an in vitro model with different blood oxygen saturation influencing the change of pulmonary artery pressure, and to provide a platform for further study on the mechanism of pulmonary hypertension.Methods: 20 New Zealand white rabbits were randomly selected as normal control group.The remaining 15 isolated pulmonary arteries were placed in culture medium and connected to the extracorporeal circulatory system. Fresh heparinized rabbit blood was perfused for 48 hours, and the perfusion pressure was maintained at 40 mm Hg(1 Hg=0.133 k Paan. The oxygen saturation of blood was controlled by adjusting the oxygen supply of the membrane lung.Three model groups were established: high saturation group (90%), middle saturation group (65%) and middle saturation group (65%), and low saturation group (40%).After bypass, the pulmonary blood vessels were obtained and compared with the normal control group. The pathological morphology, IPP6.0 quantitative analysis of pulmonary artery and the content of nitric oxide in homogenate were analyzed and compared.Results: in the three model groups, the conversion time was 48 h, the pressure was controlled at 40 mm Hg, and the blood oxygen saturation from high to low was 72.14 鹵12.85 and 43.83 鹵8.71, respectively.The results of pathological analysis showed that compared with the normal control group, the pulmonary artery wall of the three model groups was significantly thickened in vitro, and with the decrease of oxygen saturation, the elastic fibers of the pulmonary artery in the three model groups gradually increased and thickened.The difference was statistically significant (P 0.05).Conclusion: in this experiment, an isolated vascular model was established, in which different saturation of blood oxygen affected the change of pulmonary artery pressure.
