吸煙冠心病患者HDL對(duì)THP細(xì)胞膽固醇逆轉(zhuǎn)運(yùn)的影響及機(jī)制

【摘要】:目的:本研究通過(guò)提取正常健康人及吸煙冠心病患者的HDL培養(yǎng)THP-1細(xì)胞源性泡沫細(xì)胞,觀察吸煙冠心病患者的HDL對(duì)其膽固醇逆轉(zhuǎn)運(yùn)的影響及機(jī)制,為吸煙促進(jìn)動(dòng)脈粥樣硬化發(fā)病機(jī)制提供理論基礎(chǔ)。方法:采集正常健康人群及吸煙冠心病人群血樣,利用超高速離心機(jī)提取高密度脂蛋白(HDL);利用佛波酯誘導(dǎo)THP-1細(xì)胞成為巨噬細(xì)胞,然后利用氧化低密度脂蛋白(ox-LDL)誘導(dǎo)上述細(xì)胞形成泡沫細(xì)胞,并將其分為3組,分別為:空白對(duì)照組、正常人HDL組、吸煙冠心病HDL組。利用油紅O染色觀察分組泡沫細(xì)胞內(nèi)脂滴情況;利用液體閃爍計(jì)數(shù)儀從表觀現(xiàn)象方面檢測(cè)各組細(xì)胞相應(yīng)的膽固醇流出率;采用Western blot檢測(cè)以上3組三磷酸腺苷結(jié)合盒轉(zhuǎn)運(yùn)體A1(ABCA1)、三磷酸腺苷結(jié)合轉(zhuǎn)運(yùn)體G1(ABCG1)、B類Ⅰ型清道夫受體(SR-BI)相應(yīng)蛋白質(zhì)含量。結(jié)果:⑴成功提取人HDL,并鑒定;⑵佛波酯成功誘導(dǎo)THP-1細(xì)胞形成泡沫細(xì)胞,ox-LDL成功誘導(dǎo)泡沫細(xì)胞形成;⑶和空白對(duì)照組相比,正常人HDL及吸煙冠心病HDL組均可使膽固醇流出率增加(P0.05),正常人HDL組作用較吸煙冠心病HDL組作用顯著(P0.05);⑷與空白對(duì)照組相比,正常人HDL組ABCA1、ABCG1蛋白表達(dá)無(wú)明顯變化,吸煙冠心病HDL組ABCA1、ABCG1蛋白表達(dá)顯著減少(P0.05);⑸與空白對(duì)照組相比,正常人HDL組及吸煙冠心病HDL組SR-B1蛋白表達(dá)顯著升高,正常人HDL組作用較吸煙冠心病HDL組作用顯著(P0.05)。結(jié)論:正常人HDL可以促進(jìn)THP-1細(xì)胞源性泡沫細(xì)胞膽固醇逆轉(zhuǎn)運(yùn)。其可以促進(jìn)膽固醇流出,且可以升高SR-B1蛋白表達(dá),但是其并不影響ABCA1、ABCG1的蛋白表達(dá)量,而吸煙冠心病人HDL相比正常人HDL,促進(jìn)膽固醇流出作用及升高SR-B1蛋白表達(dá)作用減弱,且可以顯著減低ABCA1、ABCG1蛋白表達(dá)量,為吸煙促進(jìn)動(dòng)AS發(fā)病機(jī)制提供理論基礎(chǔ),從而減弱或者避免AS發(fā)生和發(fā)展,為冠狀動(dòng)脈粥樣硬化性心臟病的臨床預(yù)防和治療提供理論依據(jù)。
[Abstract]:Objective: to investigate the effect of HDL on cholesterol reverse transport and its mechanism by extracting HDL from healthy subjects and smoking patients with coronary heart disease (CHD) to culture THP-1 cell-derived foam cells.To provide a theoretical basis for smoking to promote the pathogenesis of atherosclerosis.Methods: the blood samples of healthy and smoking patients with coronary heart disease were collected, high density lipoprotein (HDL) was extracted by ultrahigh speed centrifuge, and THP-1 cells were induced into macrophages by phorbol ester.Then the cells were induced to form foam cells by oxidized low density lipoprotein (ox-LDL) and divided into three groups: blank control group, normal HDL group, smoking coronary heart disease HDL group.Lipid droplets in foam cells were observed by oil red O staining and cholesterol efflux was detected by liquid scintillation counter.The protein contents of adenosine triphosphate binding cassette transporter A _ (1) ABCA _ (1) and adenosine triphosphate binding transporter G _ (1) ABCG _ (1) B type 鈪,
