利拉魯肽對大鼠心肌缺血再灌注損傷的作用研究

【摘要】:目的:研究利拉魯肽對大鼠心肌缺血再灌注損傷的作用,進(jìn)一步探討其可能介導(dǎo)的機(jī)制。方法:SD大鼠48只,雄性,隨機(jī)數(shù)字表法分為假手術(shù)組、缺血再灌注組(I/R組)和利拉魯肽組。通過結(jié)扎左冠狀動脈前降支(LAD)30 min再灌注2 h建立大鼠心肌缺血再灌注損傷的實(shí)驗(yàn)?zāi)P汀LK木素伊紅染色法光鏡下觀察大鼠心肌組織的病理形態(tài)學(xué)改變;2,3,5-三苯基氯化四氮唑染色法測定各組大鼠心肌梗死面積;免疫組織化學(xué)法測定心肌組織中半胱氨酸天冬氨酸蛋白酶-3(caspase-3)和P53的表達(dá);黃嘌呤氧化酶法檢測血清超氧化物歧化酶(SOD)活性,硫代巴比妥酸法測定丙二醛(MDA)濃度。結(jié)果:蘇木素伊紅染色結(jié)果示利拉魯肽組大鼠心肌損傷程度明顯較I/R組輕。與I/R組相比,利拉魯肽組大鼠的心肌梗死面積較小[(0.44±0.08)%比(0.62±0.08)%,P0.05)],caspase-3表達(dá)水平較低(0.19±0.03比0.24±0.02,P0.05),p53的表達(dá)水平較低(0.27±0.03比0.39±0.04,P0.05),血清SOD活性較高(74.20±11.10比44.04±14.30,P0.05),MDA濃度較低(4.41±1.07比8.72±2.20,P0.05)。結(jié)論:利拉魯肽可對抗大鼠心肌缺血再灌注損傷,對心肌具有保護(hù)作用,其機(jī)制可能由抗細(xì)胞凋亡和抗氧化應(yīng)激作用介導(dǎo)。
[Abstract]:Objective: to study the effect of liralutide on myocardial ischemia-reperfusion injury in rats and to explore its possible mechanism. Methods: Forty-eight male SD rats were randomly divided into sham-operated group. Rat models of myocardial ischemia-reperfusion injury were established by ligating left anterior descending coronary artery (LADX) for 30 min and reperfusion for 2 hours. The rat heart was observed with hematoxylin and eosin red staining under light microscope. The histopathological changes of muscle tissue of rats were determined by staining method of 5-triphenyl tetrazolium chloride (5-triphenyl tetrazolium chloride). The expression of caspase-3 and p53 in myocardial tissue was determined by immunohistochemical method, and the activity of serum superoxide dismutase (SOD) was detected by xanthine oxidase method. Results: the results of hematoxylin eosin staining showed that the degree of myocardial injury in the group of liraropeptide was significantly less than that in the group I / R, and compared with that in the group I / R, and compared with that in the group I / R, the concentration of MDAs was determined by thiobarbituric acid method. The expression level of caspase-3 was lower in the group of Lilalutide (0.44 鹵0.08% vs 0.62 鹵0.08% vs 0.62 鹵0.08% than 0.62 鹵0.08). The expression level of caspase-3 was 0.19 鹵0.03 vs 0.24 鹵0.02P0.05P0.05.The expression of p53 was lower than 0.27 鹵0.03 vs 0.39 鹵0.04P0.05A, and the activity of serum SOD was 74.20 鹵11.10 vs 44.04 鹵14.30P0.05 respectively. It may be mediated by anti-apoptosis and anti-oxidative stress.
