轉(zhuǎn)化生長因子β1基因沉默脂肪干細(xì)胞移植治療血小板減少性紫癜

【摘要】:背景:有研究表明干細(xì)胞治療為血小板減少性紫癜的治療帶來了新的希望,但對于此病的完美的治療仍存在困難。目的:探討轉(zhuǎn)化生長因子β1基因沉默脂肪干細(xì)胞移植對血小板減少紫癜小鼠療效的影響。方法:將構(gòu)建的轉(zhuǎn)化生長因子β1-siRNA質(zhì)粒轉(zhuǎn)染至小鼠脂肪干細(xì)胞中,使其低表達(dá)轉(zhuǎn)化生長因子β1。用豚鼠抗小鼠血小板血清(GP-APS)腹腔注射構(gòu)建血小板減少性紫癜模型小鼠,建模后隨機(jī)分為4組,血小板減少性紫癜模型組尾靜脈注射生理鹽水20μL,脂肪干細(xì)胞組和轉(zhuǎn)化生長因子β1沉默組分別尾靜脈注射2×106 L~(-1)的CM-Dil標(biāo)記的脂肪干細(xì)胞,轉(zhuǎn)化生長因子β1沉默脂肪干細(xì)胞懸液20μL,潑尼松組按潑尼松5 mg/kg灌胃,連續(xù)治療14 d。結(jié)果與結(jié)論:(1)免疫熒光染色及EILSA檢測:與其他3組比較,轉(zhuǎn)化生長因子β1沉默組CM-Dil標(biāo)記的陽性細(xì)胞數(shù)目,血小板計(jì)數(shù),產(chǎn)板型巨核細(xì)胞比例均升高,骨髓巨核細(xì)胞總數(shù)、PAIgG及轉(zhuǎn)化生長因子β1水平下降(P0.05);(2)結(jié)果說明,轉(zhuǎn)化生長因子β1基因沉默脂肪干細(xì)胞移植能有效降低血小板減少性紫癜小鼠轉(zhuǎn)化生長因子β1及PAIgG水平,促進(jìn)骨髓巨核細(xì)胞分化成熟,從而有效提高血小板計(jì)數(shù),對治療血小板減少性紫癜有效果較好。
[Abstract]:Background: some studies have shown that stem cell therapy has brought new hope for the treatment of thrombocytopenic purpura. But it is still difficult to perfect the treatment of this disease. Objective: to investigate the effect of transforming growth factor 尾 1 gene silencing adipose stem cell transplantation on the therapeutic effect of thrombocytopenic purpura mice. Methods: transforming growth factor 尾 1-siRNA was constructed in mice with thrombocytopenic purpura. The plasmid was transfected into the adipose stem cells of mice. The mice with thrombocytopenic purpura were established by intraperitoneal injection of GP-APS. The mice were randomly divided into 4 groups after modeling. In the thrombocytopenic purpura model group, 20 渭 L saline was injected into the tail vein, and the adipose stem cells group and the transforming growth factor 尾 1 silencing group were injected with CM-Dil labeled adipose stem cells respectively. Transforming growth factor 尾 1 silenced adipose stem cell suspension (20 渭 L). Prednisone group was treated with prednisone for 5 mg/kg for 14 days. Results and conclusion Immunofluorescence staining and EILSA detection: compared with the other three groups. In TGF- 尾 1 silencing group, the number of CM-Dil labeled positive cells, the platelet count, the percentage of platelet-producing megakaryocytes increased, and the total number of bone marrow megakaryocytes (PAIgG) and the level of transforming growth factor 尾 1 (TGF- 尾 1) decreased (P 0.05). Transforming growth factor 尾 1 gene silencing adipose stem cell transplantation can effectively reduce the levels of transforming growth factor 尾 1 and PAIgG in mice with thrombocytopenic purpura, promote the differentiation and maturation of bone marrow megakaryocyte, thus effectively increase the platelet count. It has good effect on treatment of thrombocytopenic purpura.
