【摘要】：血小板活化是生理性止血中的關(guān)鍵步驟,在病理性血栓形成中也起著重要的作用。通過抑制血小板活化以預(yù)防和治療血栓是血栓性疾病防治的重要手段之一。然而,目前臨床抗血栓藥物的高效性以出血風(fēng)險(xiǎn)增加為代價(jià)。血小板糖蛋白受體VI(GPVI)是血小板特異性受體,其與膠原的結(jié)合是血小板活化的關(guān)鍵。它作為一種新型抗血栓藥物靶點(diǎn),其拮抗劑能抑制血栓及其炎癥反應(yīng),又不干擾正常止血機(jī)制,既安全又有效。本文對(duì)血小板GPVI的結(jié)構(gòu)與功能,在出血和血栓形成中的作用及其拮抗劑的最新研究進(jìn)展作一綜述,并對(duì)未來開發(fā)出具有臨床意義的以GPVI為靶點(diǎn)的抗血小板藥物進(jìn)行展望,以期為血栓栓塞性疾病提供可靠的藥物治療。
[Abstract]:Platelet activation is a key step in physiological hemostasis. It also plays an important role in the formation of pathological thrombus. Preventing and treating thrombus by inhibiting platelet activation is one of the important methods to prevent and treat thrombotic diseases. At present, the high efficiency of clinical antithrombotic drugs is at the expense of increased risk of bleeding. Platelet glycoprotein receptor VI- GPVI is a platelet-specific receptor. As a new antithrombotic target, its antagonist can inhibit thrombus and its inflammatory reaction without interfering with the normal hemostatic mechanism. In this review, the structure and function of platelet GPVI, its role in bleeding and thrombosis and its antagonist were reviewed. In order to provide reliable drug therapy for thromboembolic diseases, the future development of antiplatelet drugs with clinical significance with GPVI as the target is prospected.
