α-硫辛酸對慢性心力衰竭大鼠心功能的影響

【摘要】:目的探討α-硫辛酸(α-lipoic acid)抗氧化作用及抗心肌細胞凋亡對慢性心力衰竭大鼠心功能的影響。方法用急性心肌梗死(AMI)后大鼠心功能異常的方法構(gòu)建慢性心力衰竭(CHF)模型,6周齡的雄性SD大鼠隨機分為α-硫辛酸處理組(AMI+α-硫辛酸,n=15)、對照組(AMI+NS,n=14)和假手術(shù)組(sham-operated+NS,n=10)。術(shù)后2周后α-硫辛酸處理組給予硫辛酸(60mg/kg/d)腹腔注射,其余組大鼠等量生理鹽水(NS)腹腔注射。4周后,采用超聲心動圖分析各組大鼠的心功能、ELISA法檢測血漿腦鈉尿肽(BNP)含量、超氧化物歧化酶(SOD)、總抗氧化能力(TAC)、丙二醛(MDA)含量、HE染色觀察心肌細胞形態(tài)、免疫組化法分析心肌誘導型氧化亞氮合酶(i NOS)的表達、Western blot法測大鼠心肌組織c-Jun氨基末端激酶(JNK)及Caspase-12蛋白含量。結(jié)果對照組與假手術(shù)組大鼠相比,大鼠心功能明顯降低(射血分數(shù)由90.16%降至66.84%),血清BNP、MDA水平明顯升高(P0.05),SOD、TAC水平降低(P0.05);HE染色顯示心肌組織明顯水腫、排列紊亂,iNOS表達升高(P0.05),證明CHF建模成功;α-硫辛酸處理組與對照組相比,大鼠的心功能明顯改善(射血分數(shù)由66.84%升至80.15%),BNP、MDA含量、iNOS、JNK、Caspase-12蛋白表達均下降(P0.05),TAC、ROS水平升高(P0.05)。結(jié)論CHF建模成功;α-硫辛酸可通過抗氧化和減少細胞凋亡的方式改善CHF大鼠的心功能。
[Abstract]:Objective to investigate the effects of alpha lipoic acid (alpha -lipoic acid) on the cardiac function of rats with chronic heart failure (CHF) and the antioxidation of alpha lipoic acid (alpha acid). Methods a model of chronic heart failure (CHF) was established after cardiac dysfunction in rats with acute myocardial infarction (AMI). 6 week old male SD rats were randomly divided into alpha lipoic acid treatment group (AMI+ alpha lipoic acid, n=15), control group (AMI+NS, n=14) and sham operation group (sham-operated+NS, n=10). 2 weeks after operation, alpha lipoic acid treatment group was given intraperitoneal injection of lipoic acid (60mg/kg/d), and the rest of the rats were injected with equal amount of normal saline (NS). After 4 weeks, the echocardiography analysis of rats cardiac function, ELISA method for detection of plasma brain natriuretic peptide (BNP) content, superoxide dismutase (SOD), total antioxidant capacity (TAC), malondialdehyde (MDA) content, HE staining was used to observe the myocardial cell morphology and immunohistochemical analysis of myocardium inducible synthase (I NOS) Nitrous Oxide c-Jun N-terminal kinase measured in myocardial tissue of rats and the expression of Western blot (JNK) and Caspase-12 protein content. Results compared with control group and sham operation group rats, the heart function of rats decreased significantly (ejection fraction decreased from 90.16% to 66.84%), serum BNP and MDA levels were significantly increased (P0.05), SOD and TAC were decreased (P0.05); HE staining of myocardial tissue edema, disorder, increase the expression of iNOS (P0.05) CHF, proved successful modeling; compared with alpha lipoic acid treatment group and control group, the heart function of rats significantly improved (ejection fraction from 66.84% to 80.15%), the expression of BNP, MDA, iNOS, JNK content and Caspase-12 protein were decreased (P0.05), elevated TAC and ROS levels (P0.05). Conclusion CHF is successful in modeling, and alpha lipoic acid can improve the cardiac function of CHF rats by antioxidation and reduction of apoptosis.
