奧曲肽對(duì)肝硬化門(mén)脈高壓大鼠肝臟血紅素氧合酶-1表達(dá)的影響

目的:肝硬化(hepatic cirrhosis)是各種慢性肝病發(fā)展的晚期階段,是各種因素長(zhǎng)期反復(fù)作用導(dǎo)致了肝細(xì)胞損傷,并引起肝星狀細(xì)胞的活化,同時(shí)氧化應(yīng)激以及多種細(xì)胞因子也參與了本病的發(fā)生發(fā)展過(guò)程。門(mén)靜脈高壓癥(portal hypertension,PHT)是肝硬化失代償期主要的臨床表現(xiàn)之一,其引起的主要并發(fā)癥食管胃底靜脈曲張破裂出血嚴(yán)重威脅著患者的生命。血紅素氧合酶(heme oxygenase, HO)是血紅素代謝途徑中的起始酶和限速酶,共有3種亞型,其中HO-1活性最高,廣泛分布于全身組織,可被多種引起細(xì)胞氧化應(yīng)激的損傷因素所誘導(dǎo),HO-1及其催化產(chǎn)物膽綠素(biliverdin)、膽紅素(biliubin)和CO(carbon monoxide,CO)在體內(nèi)均有顯著的抗炎、抗細(xì)胞增殖及調(diào)節(jié)細(xì)胞因子表達(dá)等重要作用。目前,奧曲肽(octreotide, OCT)作為臨床上常用藥物治療食管胃底靜脈曲張破裂出血,但奧曲肽在門(mén)脈高壓階段的作用機(jī)制尚不完全清楚。本研究觀察奧曲肽降低門(mén)脈高壓的作用是否調(diào)控了HO-CO系統(tǒng),以探討奧曲肽降門(mén)脈高壓的可能機(jī)制,并進(jìn)一步證實(shí)HO-CO系統(tǒng)在肝硬...
