　　背景：重癥急性胰腺炎（SAP）作為一種常見的急腹癥，病因復(fù)雜，病情進(jìn)展快，可迅速發(fā)展為全身炎癥綜合癥（SIRS），進(jìn)而發(fā)展為多器官功能障礙綜合征（MODS），而內(nèi)皮細(xì)胞作為血管的最內(nèi)層屏障，其細(xì)胞間通透性增加可能參與了MODS的發(fā)生。細(xì)胞間緊密連接蛋白是細(xì)胞間連接的重要組成部分，在維持內(nèi)皮屏障完整中發(fā)揮著重要的調(diào)節(jié)作用。近年來(lái)連續(xù)性血液凈化（CBP）治療已廣泛應(yīng)用臨床搶救包括SAP、MODS在內(nèi)的各種危重患者的治療，明顯的改善了患者的預(yù)后。我們既往的研究顯示CBP治療可以改善SAP患者內(nèi)皮細(xì)胞通透性，但其機(jī)制是否與細(xì)胞間緊密連接異常表達(dá)與分布有關(guān)，有待進(jìn)一步深入研究。本文旨在探討細(xì)胞間緊密連接蛋白在CBP改善SAP患者內(nèi)皮細(xì)胞屏障功能障礙中的作用。方法：用SAP患者CBP治療的不同時(shí)間點(diǎn)血清刺激人臍靜脈內(nèi)皮細(xì)胞，或予不同濃度Rho激酶（ROCK）抑制劑Y-27632（5μ M、10μ M、20μ M）預(yù)處理后加入SAP治療前血清干預(yù)內(nèi)皮細(xì)胞。Fluorescence reader檢測(cè)單層內(nèi)皮細(xì)胞對(duì)熒光素標(biāo)記的牛血清白蛋白的通透量，免疫熒光染色檢測(cè)緊密連接蛋白Occludin、Claud...
