美洲大蠊提取物對(duì)肝星狀細(xì)胞MAPK信號(hào)通路的影響及意義的研究

目的:研究美洲大蠊提取物對(duì)大鼠肝星狀細(xì)胞(HSC-T6)MAPK信號(hào)通路中P38、P-P38、ERK1/2、P-ERK1/2、JNK和P-JNK重要信號(hào)蛋白分子表達(dá)的影響,并探討阻斷P38、ERK1/2、JNK信號(hào)通路后對(duì)HSC-T6細(xì)胞增殖的影響,旨在從信號(hào)通路的角度對(duì)美洲大蠊物提取物抗肝纖維化的相關(guān)分子機(jī)制進(jìn)行闡述。方法:1、體外培養(yǎng)大鼠肝星狀細(xì)胞(HSC-T6),用四甲基偶氮唑鹽(MTT)法檢測不同濃度(0.5 mg/mL、lmg/mL、2mg/mL、4 mg/mL、6mg/mL、8 mg/mL、10mg/mL 16mg/mL)的美洲大蠊提取物對(duì)HSC-T6細(xì)胞作用24h時(shí)的增殖情況,研究美洲大蠊提取物對(duì)HSC-T6細(xì)胞增殖的影響。2、將培養(yǎng)的對(duì)數(shù)生長期大鼠肝星狀細(xì)胞(HSC-T6)分為5組:空白對(duì)照組;濃度分別為2mg/mL、4mg/mL、6mg/mL、8mg/mL的美洲大蠊提取物實(shí)驗(yàn)組;HSC-T6經(jīng)美洲大蠊提取物作用30分鐘后,裂解細(xì)胞提取總蛋白,用Western Blot方法檢測MAPK信號(hào)通路中P38、p-P38、ERK、p-ERK、JNK、p-JNK重要信號(hào)蛋白分子表...
