β-arrestin-1調(diào)控原發(fā)性膽汁性肝硬化患者自身反應(yīng)性T細(xì)胞功能

原發(fā)性膽汁性肝硬化(PBC)的本質(zhì)是自身免疫性疾病。β-arrestin-1是一種廣泛參與了細(xì)胞內(nèi)信號(hào)轉(zhuǎn)導(dǎo)的分子,研究表明其參與了天然免疫以及獲得性免疫的調(diào)控。本課題旨在研究β-arrestin-1在PBC發(fā)病機(jī)制中的作用。通過(guò)檢測(cè)PBC患者外周血單個(gè)核細(xì)胞(PBMC)以及各淋巴細(xì)胞亞群內(nèi)β-arrestin-1的表達(dá),我們發(fā)現(xiàn)PBC患者PBC內(nèi)β-arrestin-1的表達(dá)在蛋白水平和mRNA水平較健康個(gè)體以及乙型肝炎后肝硬化患者顯著增高,且與患者的Mayo危險(xiǎn)評(píng)分呈正相關(guān)。在自身抗原的刺激下,β-arrestin-1可以促進(jìn)PBC患者自身反應(yīng)性T細(xì)胞的増殖、增加IFN-γ的表達(dá)、降低細(xì)胞內(nèi)轉(zhuǎn)錄因子NF-κB和AP-1的活性。此外,還可以促進(jìn)自身免疫相關(guān)基因CD40L,LIGHT,IL-17和IFN-γ的表達(dá),抑制TRAIL,Apo2和HDAC7A的表達(dá)。進(jìn)一步的研究發(fā)現(xiàn)β-arrestin-1可以調(diào)節(jié)上述7個(gè)與自身免疫相關(guān)的基因啟動(dòng)子區(qū)域的乙;健1狙芯勘砻:β-arrestin-1通過(guò)表觀遺傳學(xué)機(jī)制調(diào)節(jié)PBC患者自身反應(yīng)性T細(xì)胞的功能,可能是PBC潛在的治療靶點(diǎn)。
