MYDGF對小鼠非酒精性脂肪肝的保護(hù)作用及其機(jī)制研究

研究背景和目的骨髓是否調(diào)節(jié)全身代謝仍然未知。我們的最新研究表明,由骨髓細(xì)胞(bone marrow cells,BMCs)分泌的髓源性生長因子(myeloid-derived growth factor,MYDGF)能夠減輕2型糖尿病小鼠的胰島素抵抗。在這里,我們探討MYDGF是否能改善非酒精性脂肪性肝。╪on-alcoholic fatty liver disease,NAFLD),并作為骨髓與肝臟之間的信使分子來調(diào)節(jié)肝臟脂肪代謝。方法1、骨髓細(xì)胞特異性MYDGF敲除小鼠(MYDGF-/-,KO)和野生型對照(MYDGF+/+,WT)小鼠接受高脂飲食(high-fat diet,HFD)喂養(yǎng),從而建立NAFLD模型。2、通過尾靜脈注射腺相關(guān)病毒(adeno-associated viruses,AAV)-MYDGF在肝臟中超表達(dá)MYDGF,通過骨髓移植(bone marrow transplantion,BMT)和骨髓腔內(nèi)原位注射AAV-MYDGF在骨髓中超表達(dá)MYDGF。通過siRNA沉默骨髓細(xì)胞特異性MYDGF敲除小鼠肝臟的IKKβ。3、采用蘇木素-伊紅染色、油紅O染色和電子顯...
