Cajal樣間質(zhì)細(xì)胞調(diào)控oddi括約肌運(yùn)動(dòng)及在oddi括約肌功能障礙發(fā)病中的作用

背景自1887年Oddi括約。╯phincter of Oddi,SO)被發(fā)現(xiàn)以來(lái),人們對(duì)其結(jié)構(gòu)及功能的認(rèn)識(shí)也不斷深入,且清楚地認(rèn)識(shí)到其對(duì)膽胰正常功能的維持具有重要的作用。我們?cè)谇捌诘囊幌盗醒芯堪l(fā)現(xiàn)SO的解剖及功能異常與多種膽胰疾病有關(guān),如原發(fā)膽管色素結(jié)石、膽囊切除術(shù)后綜合征、先天性膽胰管合流異常、急性胰腺炎及Oddi括約肌功能障礙(sphincter of Oddi dysfunction,SOD)等。由于SO特殊的解剖位置及生理功能,目前對(duì)其功能調(diào)控的基礎(chǔ)研究明顯滯后。內(nèi)鏡下SO切開(kāi)(endoscopic sphincterotomy,EST)對(duì)于SOD的治療的近期效果滿意,但是EST相關(guān)并發(fā)癥及其后腸膽反流的發(fā)生仍是臨床上難以解決的問(wèn)題。SO的自主動(dòng)力本質(zhì)上是肌源性的,存在多種神經(jīng)以調(diào)節(jié)其運(yùn)動(dòng),同時(shí)協(xié)調(diào)SO運(yùn)動(dòng)與十二指腸,膽囊和胃的運(yùn)動(dòng)。SO動(dòng)力的改變可能是來(lái)自于神經(jīng)和生物活性物質(zhì)在平滑肌水平上的信號(hào)的整合,SO中興奮的起源和傳導(dǎo)機(jī)制仍不清楚。已描述在胃腸道中存在產(chǎn)生擺動(dòng)性電活動(dòng)的特殊的起搏細(xì)胞,即Cajal樣細(xì)胞(interstitial Cajal-like cells,I...

.5.2免疫熒光化學(xué)染色實(shí)驗(yàn)分組及
中國(guó)醫(yī)科大學(xué)博士學(xué)位論文6遠(yuǎn)端,取下完整壺腹部標(biāo)本(如圖1)。 迅速放入磷酸鹽緩沖液(PBS) 中。用PBS充分沖洗膽道和十二指腸, 然后置于4%多聚甲醛溶液中固定48h(根據(jù)組織大小和致密程度調(diào)整固定時(shí)間),取出后,再放入20%蔗糖中24h、30%蔗糖中48h脫水。將組織塊修剪后放入冷凍包埋模具中,調(diào)整切面位置,加入包埋劑,置于恒冷箱冰凍切片機(jī)切片架上,待組織塊冷凍變硬后切片,厚度為8~12μm。將切片貼附在處理過(guò)的防脫載玻片上,室溫干燥1小時(shí)后,放入載片盒,-80°C密閉保存?zhèn)溆谩H〕霰鶅銮衅,?fù)溫30min,以4%多聚甲醛固定30min, PBS 漂洗5min *3 次, 以5% 胎牛血清處理標(biāo)本1h
