自噬和長(zhǎng)鏈非編碼RNA MEG3對(duì)IGFBPrP1激活肝星狀細(xì)胞的調(diào)控及其機(jī)制的研究

研究背景:肝星狀細(xì)胞(hepatic stellate cells,HSCs)是來(lái)源于間質(zhì)的一種肝臟非實(shí)質(zhì)細(xì)胞,HSCs的激活是引起肝纖維化發(fā)生的關(guān)鍵環(huán)節(jié)。多種細(xì)胞因子參與其活化,其中轉(zhuǎn)化生長(zhǎng)因子(transforming growth factor beta1,TGFβ1)發(fā)揮著舉足輕重的作用,在活化的初期及末期通過(guò)直接或間接方式持續(xù)激活HSCs,HSCs激活后可使肝臟內(nèi)細(xì)胞外基質(zhì)(extracellular matrix,ECM)合成和降解失衡,最終導(dǎo)致肝纖維化。細(xì)胞自噬是指細(xì)胞在各種因素的作用下,細(xì)胞對(duì)其內(nèi)部受損的細(xì)胞器、錯(cuò)誤折疊的蛋白質(zhì)和侵入的病原體進(jìn)行降解的生物學(xué)過(guò)程,F(xiàn)已證實(shí),自噬在肝纖維化中發(fā)揮重要作用。長(zhǎng)鏈非編碼RNA(long noncoding RNA,lncRNA)是一種轉(zhuǎn)錄長(zhǎng)度超過(guò)200核苷酸的RNA,母系表達(dá)基因3(materally expressed gene 3,MEG3),是最先發(fā)現(xiàn)的具有腫瘤抑制功能的lncRNA,其在肝癌組織中的表達(dá)量低于正常組織約200倍,且在肝纖維化中也發(fā)揮重要作用。胰島素樣生長(zhǎng)因子結(jié)合蛋白相關(guān)蛋白1(insulin-like ...

新鮮分離的大鼠原代HSCs×200
