樹(shù)鼩慢性乙型肝炎病毒感染模型的探索

HBV感染人體以后可以造成急性肝炎、慢性肝炎,可發(fā)展為肝硬化,甚至導(dǎo)致肝細(xì)胞癌變,嚴(yán)重影響人類(lèi)生命健康。雖然新生兒乙肝疫苗接種計(jì)劃有效降低了新發(fā)HBV感染的發(fā)生率,但對(duì)于全世界已有的龐大HBV慢性感染人群仍缺乏徹底、有效的治療措施。對(duì)HBV感染致病機(jī)制的研究及防治藥物的研發(fā)離不開(kāi)合適的動(dòng)物感染模型。長(zhǎng)期以來(lái),黑猩猩是唯一可以被用于HBV感染研究的易感動(dòng)物。然而,黑猩猩屬于瀕危珍稀動(dòng)物、來(lái)源受限,其相對(duì)大型的體格、昂貴的使用成本,以及受到倫理因素影響,黑猩猩作為實(shí)驗(yàn)動(dòng)物的使用越來(lái)越受到限制。因此,人們迫切需要尋找其他更加方便、穩(wěn)定的小型動(dòng)物作為模型以利于HBV研究的開(kāi)展。由于其嚴(yán)格的宿主特異性,HBV感染難以在除人類(lèi)和黑猩猩以外的其他動(dòng)物實(shí)現(xiàn),很多關(guān)于HBV的認(rèn)識(shí)都是通過(guò)對(duì)與HBV相似的其他病毒的研究獲得的。鴨乙型肝炎病毒(Duck hepatitis B virus,DHBV)和土撥鼠肝炎病毒(Woodchuck hepatitisvirus,WHV)與HBV同屬嗜肝DNA病毒,它們?cè)诟髯运拗黧w內(nèi)的感染為人們了解屬嗜肝DNA病毒的感染復(fù)制周期及宿主免疫系統(tǒng)反應(yīng)提供了大量依據(jù)。但是鴨和土...
