目的研究慢性乙型肝炎（CHB）患者血清中HBcAg以什么方式存在。方法1、采用聚乙二醇（PEG）沉淀法沉淀血清樣本中的HBV特異性循環(huán)免疫復合物并用胃蛋白酶解離復合物;2、通過免疫印跡檢測解離物中的HBcAg復合物;3、用親脂染料預染解離物,通過瓊脂糖電泳檢測脂質成分的存在。結果通過PEG沉淀,胃蛋白酶解離及Western Blot確定慢性乙型肝炎患者血清中有HBcAg特異性復合物存在,進一步預染樣品后瓊脂糖電泳證明HBcAg復合物上結合有脂質成分。結論證實了慢性乙型肝炎患者血清中有HBcAg特異性免疫復合物的存在,而且HBcAg是與脂質成分結合,說明血清中HBcAg是以脂復合物的形式存在著,從而推測慢性乙型肝炎患者血清中的HBcAg結合有脂膜成分,是膜表達HBcAg的殘留,可能是免疫學系統(tǒng)攻擊的靶抗原。
