CTLA4Ig基因修飾人肝細胞株L02誘導免疫耐受的實驗研究

肝細胞移植是治療肝衰竭和代謝性肝病的重要手段之一,相對于肝臟移植,肝細胞移植具有來源較廣、費用相對低廉、創(chuàng)傷較小等優(yōu)點,可提供暫時性肝功能支持,為肝臟重建創(chuàng)造機會。但同肝移植一樣,排斥反應影響移植肝細胞的長期存活和遠期療效。目前解決排斥反應的主要策略是使用免疫抑制劑,但此類藥物毒副作用較大,需長期甚至終身使用,且增加感染和腫瘤的發(fā)生率。在此背景下,誘導移植物免疫耐受成為研究熱點。細胞毒性T淋巴細胞相關抗原4免疫球蛋白(cytotoxic T-lymphocyte associated antigen 4-immunoglobulin, CTLA4Ig)是CTLA4分子胞外功能區(qū)與免疫球蛋白IgG恒定區(qū)相結合的融合性可溶性蛋白,可與T淋巴細胞表面的CD28分子競爭性結合抗原遞呈細胞(antigen presenting cell, APC)表面的B7分子,阻斷T細胞活化的共刺激通路,抑制T細胞活化,誘導對特異性抗原的免疫耐受。大量實驗已經(jīng)證實CTLA4Ig可以有效抑制排斥反應,延長移植物存活時間,肝細胞移植中應用CTLA4Ig同樣被證實可有效誘導免疫耐受。其主要應用方法為CTLA4Ig蛋...
