CHI3L1調(diào)控Th1細胞分化保護急性肝損傷的機制研究

第一部分CHI3L1在硫代乙酰胺導(dǎo)致的急性肝損傷中的作用目的探討幾丁質(zhì)酶3樣蛋白1(Chitinase-3-like-1,CHI3L1)在硫代乙酰胺(thioacetamide,TAA)導(dǎo)致的急性肝損傷中的作用。方法選擇臨床中存在肝功能損傷的患者(30例)和健康體檢者(10例),生化儀檢測血清ALT、AST水平,ELISA檢測CHI3L1的水平。利用不同劑量TAA(80mg/kg、150mg/kg)在C57BL/6小鼠體內(nèi)構(gòu)建急性肝損傷模型,3天后利用qRT-PCR檢測肝內(nèi)CHI3L1mRNA的表達,檢測血清ALT、AST、CHI3L1水平。使用均為C57BL/6背景的小鼠建立3組TAA急性肝損傷模型,分別為CHI3L1基因敲除(CHI3L1-KO)組、野生型(WT)組和尾靜脈注射重組CHI3L1蛋白(rCHI3L1)組,檢測三組血清ALT、AST水平,組織病理學(xué)評價肝臟損傷程度(以Suzuki’s評分量化)。qRT-PCR技術(shù)檢測肝內(nèi)IFN-γ、IL-4、IL-17A和Foxp3的mRNA表達,流式細胞分析技術(shù)檢測肝內(nèi)CD4+IFN-γ+、...
