銀杏總黃酮對NAFLD大鼠脂聯(lián)素信號(hào)通路的影響

【摘要】:目的探討銀杏總黃酮對非酒精性脂肪性肝病(nonalcoholic fatty liver disease,NAFLD)大鼠脂聯(lián)素信號(hào)通路的影響。方法利用高脂飼料喂養(yǎng)法構(gòu)建NAFLD大鼠模型。72只SPF級(jí)雄性大鼠分為正常對照組、NAFLD模型組和銀杏總黃酮治療組3組,每組24只。測定各組大鼠體質(zhì)量及肝指數(shù),光鏡觀察肝臟組織形態(tài)改變,RT-PCR法檢測大鼠肝臟AdipoR2mRNA、PPARαmRNA的表達(dá),Western blot法檢測各組大鼠肝臟AdipoR2、PPARα、AMPK蛋白表達(dá)以及AMPK蛋白磷酸化水平。結(jié)果 NAFLD大鼠造模成功。第4周末,與正常對照組大鼠相比,模型組及治療組大鼠體質(zhì)量與肝指數(shù)均增高(均P0.05),肝臟AdipoR2mRNA、PPARαmRNA的表達(dá)水平降低,肝臟AdipoR2、PPARα、AMPK蛋白表達(dá)以及AMPK蛋白磷酸化水平降低,差異有統(tǒng)計(jì)學(xué)意義(均P0.05);模型組與治療組相比,肝臟AdipoR2mRNA、PPARαmRNA的表達(dá)水平差異無統(tǒng)計(jì)學(xué)意義(均P0.05),肝臟AdipoR2、PPARα、AMPK蛋白表達(dá)及AMPK蛋白磷酸化水平差異無統(tǒng)計(jì)學(xué)意義(均P0.05)。第8、12周末,模型組大鼠體質(zhì)量與肝指數(shù)水平逐漸增高,肝臟AdipoR2mRNA、PPARαmRNA的表達(dá)水平,AdipoR2、PPARα、AMPK蛋白表達(dá)水平及AMPK蛋白磷酸化逐漸下降;治療組大鼠體質(zhì)量及肝指數(shù)逐漸接近正常對照組,肝臟AdipoR2mRNA、PPARαmRNA的表達(dá)水平增高(均P0.05),AdipoR2、PPARα、AMPK蛋白表達(dá)及AMPK蛋白磷酸化水平逐漸增高(均P0.05)。結(jié)論銀杏總黃酮能控制大鼠體質(zhì)量與肝指數(shù),提高大鼠肝臟AdipoR2mRNA、PPARαmRNA和AdipoR2、PPARα、AMPK蛋白表達(dá)水平,并使AMPK蛋白磷酸化水平升高。推測銀杏總黃酮可能通過增強(qiáng)脂聯(lián)素信號(hào)通路的活性,一方面減少肝細(xì)胞異常凋亡,另一方面促進(jìn)線粒體內(nèi)脂肪酸的氧化,從而減輕NAFLD的肝損傷,阻止NAFLD的進(jìn)展。
[Abstract]:Objective to investigate the effect of total flavonoids of Ginkgo biloba on adiponectin signaling pathway in (nonalcoholic fatty liver disease,NAFLD rats. Methods 72 SPF male rats were divided into normal control group, NAFLD model group and ginkgo total flavonoid treatment group with 24 rats in each group. The body mass and liver index of each group were measured. The morphologic changes of liver were observed under light microscope. The expression of AdipoR2mRNA,PPAR 偽 mRNA in rat liver was detected by RT-PCR method. The expression of AdipoR2,PPAR 偽, AMPK protein and the phosphorylation of AMPK protein were detected by, Western blot method. Results the model of NAFLD rats was successfully established. At the end of the 4th week, compared with the normal control group, the body mass and liver index of the model group and the treatment group were increased (P0.05), the expression of AdipoR2mRNA,PPAR 偽 mRNA in the liver decreased, and the expression of AdipoR2,PPAR 偽 in the liver decreased. The expression of AMPK protein and the level of phosphorylation of AMPK protein decreased significantly (P0.05). There was no significant difference in the expression of AdipoR2mRNA,PPAR 偽 mRNA between the model group and the treatment group (P0.05), but there was no significant difference in the expression of AdipoR2,PPAR 偽, AMPK protein and the phosphorylation of AMPK protein in the liver (P0.05). At the end of the 812th week, the body mass and liver index of the model group increased gradually, while the expression of AdipoR2mRNA,PPAR 偽 mRNA, AdipoR2,PPAR 偽 and AMPK protein and the phosphorylation of AMPK protein decreased gradually in the liver of the model group. In the treatment group, the body mass and liver index were close to the normal control group, the expression of AdipoR2mRNA,PPAR 偽 mRNA in the liver increased (P0.05), the expression of AdipoR2,PPAR 偽, AMPK protein and the phosphorylation of AMPK protein increased gradually (P0.05). Conclusion Total flavonoids of Ginkgo biloba can control the body mass and liver index of rats, increase the expression of AdipoR2mRNA,PPAR 偽 mRNA, AdipoR2,PPAR 偽 and AMPK protein, and increase the phosphorylation level of AMPK protein. It is speculated that the total flavonoids of Ginkgo biloba may reduce the abnormal apoptosis of hepatocytes by enhancing the activity of adiponectin signaling pathway on the one hand and promote the oxidation of fatty acids in mitochondria on the other hand thereby alleviate the liver injury of NAFLD and prevent the progress of NAFLD.
