【摘要】：目的研究美洲大蠊水提取物對免疫性肝纖維化大鼠的保護(hù)作用及部分機制。方法將SD大鼠隨機分為正常組、模型組、美洲大蠊30 mg/kg、60 mg/kg和120 mg/kg組和秋水仙堿物組,每組10只。免疫性肝纖維化模型通過大鼠腹腔注射豬血清制備,0.5 ml/kg,每周2次,共12周。造模期間給藥組分別給予相應(yīng)的受試藥物,正常組和模型組分別給予等劑量的生理鹽水。于造模給藥結(jié)束后處死大鼠,測定肝脾指數(shù);收集外周血,測定血清中丙氨酸氨基轉(zhuǎn)移酶(ALT)、門冬氨酸氨基轉(zhuǎn)移酶(AST)、透明質(zhì)酸(HA)、層粘蛋白(LN)、Ⅲ型前膠原酶(PCⅢ)和Ⅳ型膠原酶(CⅣ)含量;取肝臟,HE、Masson染色觀察肝組織病理及膠原沉積變化;制備肝臟勻漿,ELISA法測定超氧化物歧化酶(SOD)活性和丙二醛(MDA)、谷光甘肽過氧化物酶(GSH-Px)含量;免疫組織化學(xué)和Western blot法測定肝組織中α-SMA、TGF-β1蛋白的表達(dá)。結(jié)果與模型組比較,美洲大蠊(30、60、120 mg/kg)能降低肝纖維化大鼠升高的肝脾指數(shù),降低血清中ALT、AST、HA、LN、PCⅢ、CⅣ水平,降低肝組織MDA含量,提高SOD及GSH活性,減輕大鼠肝臟病理損傷程度,減少肝組織中α-SMA、TGF-β1的表達(dá)。結(jié)論美洲大蠊水提取物對豬血清誘導(dǎo)的免疫性肝纖維化大鼠有顯著的保護(hù)作用,其機制可能與其抗氧化作用及抑制TGF-β1的表達(dá)有關(guān)。
[Abstract]:Objective to study the protective effect and mechanism of water extract of Blattella Americana on immune hepatic fibrosis in rats.
