克羅恩病人腸黏膜炎性區(qū)域真菌菌群組成變化研究

【摘要】:目的 :比較克羅恩病人腸黏膜炎性與非炎性區(qū)域真菌菌群組成的差異,探討?zhàn)つふ婢鹤兓c克羅恩病的潛在聯(lián)系。方法 :手術(shù)收集7例活動性克羅恩病人腸黏膜炎性與非炎性區(qū)域組織,提取DNA,采用巢式PCR擴(kuò)增18S r RNA基因片段,行變性梯度凝膠電泳,確定腸黏膜真菌菌群多樣性和組成。結(jié)果 :與非炎性黏膜相比,炎性區(qū)域真菌菌群豐富度和多樣性顯著升高(P0.05)。腸黏膜炎性區(qū)域真菌菌群組成發(fā)生顯著改變,主要表現(xiàn)為條件致病白色念珠菌,熱帶假絲酵母菌,串珠狀赤霉菌,甘藍(lán)鏈格孢菌和新型隱球菌的相對豐度顯著增加(P0.05);而共生菌釀酒酵母菌,芽殖酵母菌,產(chǎn)黃青霉菌和雙色蠟?zāi)⒕@著降低(P0.05)。結(jié)論 :克羅恩病人腸黏膜炎性區(qū)域真菌菌群嚴(yán)重失衡,條件致病真菌定植增加可能參與了腸黏膜炎性損傷的形成。
[Abstract]:Aim: to study the relationship between mucosal fungal flora change and Crohn's disease. Methods: the intestinal mucosal inflammatory and non-inflammatory tissues were collected from 7 active Crohn patients. The 18s r RNA gene fragment was amplified by nested PCR, and the bacterial diversity and composition of intestinal mucosal fungi were determined by denaturing gradient gel electrophoresis (DGGE). Results: compared with non-inflammatory mucosa, the abundance and diversity of fungal flora in inflammatory region were significantly increased (P0.05). The composition of fungal flora in the inflammatory area of intestinal mucosa changed significantly, mainly showing that the relative abundance of Candida albicans, Candida tropicalis, Streptococcus cerevisiae, Alternaria napus and Cryptococcus neoformans increased significantly (P0.05). While symbiotic bacteria, Saccharomyces cerevisiae, Bacillus sprouts, Penicillium flavus and Tricholoma bicolor decreased significantly (P0.05). Conclusion: the fungal flora in Crohn's intestinal mucosal inflammatory region is seriously out of balance and the increased colonization of conditional pathogenic fungi may be involved in the formation of intestinal mucosal inflammatory injury.
