【摘要】：目的探討非禁食和禁食對大鼠應激性胃潰瘍胃黏膜損傷及MMP-9、TIMP-2表達的影響。方法非禁食應激組不予禁食,禁食應激組于應激前24 h禁食。用浸水-束縛應激(waterimmersion and restraint stress,WIRS)建立大鼠應激性潰瘍模型。應激3 h后,觀察非禁食應激組和禁食應激組大鼠胃黏膜病變,對潰瘍指數(shù)(UI)進行評分;組織學觀察胃出血;用Western blot檢測胃黏膜MMP-9、TIMP-2的蛋白表達。結果 WIRS應激3 h能成功誘導大鼠應激性胃潰瘍。非禁食應激組與禁食應激組比較,應激性胃潰瘍大鼠的UI后者高于前者(P0.01),黏膜層炎性細胞浸潤數(shù)后者高于前者(P0.01),WIRS大鼠胃黏膜MMP-9蛋白表達后者顯著高于前者(P0.01),而TIMP-2蛋白表達后者略低于前者(P0.05)。結論非禁食應激可顯著減少MMP-9表達。非禁食處理減輕浸水-束縛應激大鼠胃黏膜損傷。
[Abstract]:Objective to investigate the effects of non-fasting and fasting on gastric mucosal injury and MMP-9,TIMP-2 expression in rats with stress-induced gastric ulcer. Methods No fasting was observed in the non-fasting stress group, but no fasting was observed in the non-fasting stress group 24 hours before stress. Rat stress ulcer model was established by immersion-restraint stress (waterimmersion and restraint stress,WIRS). After 3 hours of stress, gastric mucosal lesion was observed in non-fasting stress group and fasting stress group, ulcer index (UI) was scored, gastric hemorrhage was observed by histology, and MMP-9,TIMP-2 protein expression in gastric mucosa was detected by Western blot. Results WIRS stress for 3 h could successfully induce stress gastric ulcer in rats. The comparison between non-fasting stress group and fasting stress group, The UI of rats with stress gastric ulcer was higher than that of the former (P0.01), and the number of inflammatory cell infiltration in the mucosal layer was higher than that of the former (P0.01), WIRS). The latter was significantly higher than that of the former (P0.01), while the expression of TIMP-2 protein was slightly lower than that of the former (P0.05). Conclusion non-fasting stress can significantly reduce the expression of MMP-9. Non-fasting treatment alleviated gastric mucosal injury in rats with immersion-restraint stress.
