荔枝核總黃酮對大鼠肝纖維化PDGF、TNF-α因子的影響

【摘要】:目的:探討荔枝核總黃酮(total flavone from Litchi Chinensis Sonn,TFL)對大鼠肝纖維化PDGF及TNF-α因子的影響。方法:SD雄性大鼠60只,,隨機分為空白組、模型組、水飛薊賓(SIL)組(50mg kg-1d-1)、TFL高劑量組(400mg kg-1d-1)、TFL中劑量組(200mg kg-1d-1)、TFL低劑量組(100mg kg-1d-1)六組。采用二甲基亞硝胺DMN腹腔注射制備SD大鼠進行肝纖維化造模,造模4周,造模同時SIL、TFL等四組灌胃給藥干預(yù),6周后處死大鼠,觀察大鼠肝纖維化水平,用放射免疫法測定模型組、SIL組和TEL組大鼠血清中層粘蛋白(LN)、透明質(zhì)酸(HA)及Ⅲ型前膠原(PCⅢ)水平,用HE染色法和Masson染色觀察大鼠肝組織鏡下病理變化纖維化水平,采用PCR法檢測SIL組和TFL組PDGF、TNF因子表達水平變化,用SPSS11.0軟件對數(shù)據(jù)進行統(tǒng)計分析。結(jié)果:與模型組大鼠比較, SIL和TFL大、中劑量治療后大鼠血清中HA、LN和PCⅢ的水平顯著降低(P<0.01);肝組織內(nèi)纖維化程度降低,肝小葉結(jié)構(gòu)趨于正常;TFL大、中劑量給藥組和水飛薊素組血清PDGF及TNF-α水平均顯著低于模型組(P<0.05);兩組大鼠肝組織PDGF及TNF-α表達明顯降低(P<0.05)。TFL中劑量給藥組與對照組比較血清HA、LN和PCⅢ的水平、肝纖維化程度、PDGF及TNF-α表達水平均無明顯差異(P>0.05)。結(jié)論:TFL可以下調(diào)二甲基亞硝胺肝纖維化大鼠的PDGF、TNF-α細胞因子表達水平,具有明顯的抗大鼠肝纖維化作用。

SD rats were induced by intraperitoneal injection of dimethylnitrosamine (DMN) to model liver fibrosis for 4 weeks. Four groups of rats, such as SIL,TFL, were given intragastric administration of dimethylnitrosamine. After 6 weeks, the rats were killed to observe the level of hepatic fibrosis.
