【摘要】：目的探討抵抗素在伴高脂血癥性急性壞死性胰腺炎(ANP)發(fā)病機(jī)制中的可能作用。方法將50只雄性SD大鼠分為正常對(duì)照組(C組)5只,單純ANP組(ANP組)15只、高脂對(duì)照組(H組)15只及高脂ANP組(HAP組)15只,分組采用高脂飼料喂養(yǎng)4周建立大鼠高脂血癥模型,應(yīng)用逆行胰膽管3.5%�；悄懰徕c注射構(gòu)建ANP大鼠模型,分別于造模后4,9,24h各處死5只大鼠。ELISA法測定各組不同時(shí)段血清抵抗素和TNF-α水平,臨床全自動(dòng)生化儀檢測血脂及血清淀粉酶(Amyl),免疫組織化學(xué)方法觀察胰腺組織中抵抗素及NF-κB p65表達(dá),光鏡下觀察胰腺組織病理改變。結(jié)果 HAP組較ANP組胰腺炎病理損傷加重(病理評(píng)分P0.05)。造模后各時(shí)間點(diǎn),HAP組較ANP組血清TNF-α水平(P0.05)和抵抗素水平(P0.01)更高,胰腺組織NF-κB p65表達(dá)水平(P0.05)和抵抗素表達(dá)水平(P0.05)更高。血清TNF-α和抵抗素水平呈正相關(guān)(r=0.698,P0.05);胰腺組織NF-κB p65表達(dá)和抵抗素表達(dá)呈正相關(guān)(r=0.839,P0.01)。結(jié)論在ANP發(fā)病中,抵抗素可能通過促進(jìn)轉(zhuǎn)錄因子NF-κB p65的表達(dá)及TNF-α的釋放發(fā)揮生物學(xué)效應(yīng);HAP胰腺組織病理損害較重可能與抵抗素表達(dá)的增加相關(guān)。
[Abstract]:Objective to investigate the possible role of resistin in the pathogenesis of (ANP) with hyperlipidemic acute necrotizing pancreatitis. Methods Fifty male SD rats were divided into normal control group (group C, n = 5), ANP group (ANP group, n = 15), hyperlipidemia group (H group, n = 15) and hyperlipidemia group (HAP group, n = 15). The rat model of ANP was established by injecting 3.5% sodium taurocholate into retrograde pancreaticobiliary duct. Five rats were killed at 9 h after the establishment of the model. The levels of resistin and TNF- 偽 in serum of each group were measured by ELISA method at different time points. The expression of resistin and NF- 魏 B p65 in pancreatic tissue was detected by immunohistochemical method with serum amylase (Amyl), and the histopathological changes of pancreatic tissue were observed under light microscope. Results the pathological injury in HAP group was more serious than that in ANP group (P 0.05). The levels of serum TNF- 偽 (P0.05) and resistin (P0.01) in HAP group were higher than those in ANP group. The expression levels of NF- 魏 B p65 and resistin in pancreatic tissue were higher than those in ANP group (P0.05). There was a positive correlation between serum TNF- 偽 and resistin levels (P 0.05), and a positive correlation between the expression of NF- 魏 B p65 and resistin expression in pancreatic tissues (r = 0.839, P 0.01). Conclusion resistin may play a biological role in the pathogenesis of ANP by promoting the expression of transcription factor NF- 魏 B p65 and the release of TNF- 偽, and the histopathological damage of HAP pancreas may be related to the increase of resistin expression.
