利用基質(zhì)輔助激光解析電離飛行時(shí)間串聯(lián)質(zhì)譜分析膽囊結(jié)石中非黏性蛋白的組成

【摘要】:目的探討膽固醇結(jié)石和膽色素結(jié)石中是否存在非黏性蛋白并鑒定蛋白質(zhì)的組成。方法選取30例行保膽取石術(shù)患者的膽囊結(jié)石,男性11例,女性19例。采用肉眼觀察并結(jié)合三酰甘油檢測(cè)試劑盒(GPO-PAP)測(cè)定膽固醇含量的方法對(duì)膽結(jié)石進(jìn)行分類(lèi),分為20例膽固醇型和10例膽色素型。應(yīng)用組織裂解法(RIPA)提取膽囊結(jié)石蛋白,并用十二烷基硫酸鈉聚丙烯酰胺凝膠電泳(SDS-PAGE)與基質(zhì)輔助激光解析電離飛行時(shí)間串聯(lián)質(zhì)譜(MALDI-TOF-TOF)分析相結(jié)合的方法對(duì)該蛋白進(jìn)行鑒定分析。通過(guò)DAVID工具對(duì)上述基因進(jìn)行基因本體(GO)功能富集分析本圖及京都基因、基因組百科全書(shū)(KEGG)信號(hào)轉(zhuǎn)導(dǎo)通路富集分析。結(jié)果膽固醇結(jié)石和膽色素結(jié)石中出現(xiàn)特異性蛋白,經(jīng)質(zhì)譜鑒定為轉(zhuǎn)鐵蛋白前體蛋白(TRFE)、白蛋白(ALB)、抗胰蛋白酶1(SERPINA1)、免疫球蛋白γ-1(IGHG1)、α血紅蛋白(HBA1)、β血紅蛋白(HBB),其中IGHG1不存在于膽色素結(jié)石中。結(jié)論 TRFE、ALB、SERPINA1、HBA1、HBB等蛋白共同存在于膽固醇和膽色素結(jié)石中,而IGHG1只存在于膽固醇結(jié)石中,可見(jiàn)IGHG1蛋白或其對(duì)應(yīng)基因是參與不同膽結(jié)石類(lèi)型成因的重要元素及生物標(biāo)志物。KEGG分析結(jié)果表明,SERPINA1參與補(bǔ)體和凝血級(jí)聯(lián)反應(yīng)。鑒定和分析膽囊結(jié)石中非黏性蛋白的成分及利用資料庫(kù)的模擬對(duì)闡明膽囊結(jié)石的發(fā)病機(jī)制有重要的意義。
[Abstract]:Objective to investigate the presence of non-viscosity protein in cholesterol gallstone and pigment gallstone and to identify the protein composition. Methods 30 patients (11 males and 19 females) underwent cholecystolithiasis. Gallstones were classified into 20 cholesterol type and 10 bile pigment type by naked eye observation and triglyceride detection kit (GPO-PAP). Gallstone protein was extracted by tissue lysis method (RIPA) and identified by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) combined with matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-TOF). Analysis of gene bulk (GO) function enrichment by DAVID tools. This map and Kyoto gene, genome encyclopedia (KEGG) signal transduction pathway enrichment analysis. Results specific proteins were found in cholesterol gallstones and pigment gallstones, which were identified as transferrin precursor protein (TRFE), albumin (ALB), antitrypsin 1 (SERPINA1), immunoglobulin 緯 -1 (IGHG1), 偽 hemoglobin (HBA1) and 尾 hemoglobin (HBB), in which IGHG1 was not present in gallstones. Conclusion TRFE,ALB,SERPINA1,HBA1,HBB and other proteins co-exist in cholesterol and pigment gallstone, while IGHG1 exists only in cholesterol stone. IGHG1 protein or its corresponding gene is an important element and biomarker. KEGG analysis shows that SERPINA1 is involved in complement and coagulation cascade reaction. It is important to identify and analyze the components of non-mucin in gallstone and use the simulation of database to elucidate the pathogenesis of gallstone.
