【摘要】：目的探索并建立一種基于循環(huán)micro RNA的診斷大鼠潰瘍性結(jié)腸炎(ulcerative colitis,UC)方法并監(jiān)測其治療后效果的臨床前策略。方法第1階段:硫酸葡聚糖(Dextran sulfate sodium,DSS)誘導(dǎo)大鼠UC模型。實(shí)時(shí)定量RT-PCR篩選差異性的血清micro RNA。ELISA檢測血清核周抗中性粒細(xì)胞胞漿抗體(periunclear antineutrophil cytoplasmic antibodies,p ANCA)。Logistic回歸法建立診斷模型Indice。受試者工作特征(receiver operating characteristic,ROC)曲線評價(jià)指標(biāo)的診斷效能。第2階段:驗(yàn)證指標(biāo)對UC的診斷效能,并用其評價(jià)UC的療效。結(jié)果第1階段:大鼠UC模型建立成功。Mi R-20b、mi R-21和let-7e*在UC大鼠血清中高表達(dá),mi R-125b-1*無明顯變化。p ANCA在UC大鼠血清中高表達(dá)。診斷模型Indice=0+(2.645×mi R-20b)+(1.622×mi R21)+(0.979×let-7e*)+(0.082×p ANCA)的診斷效能強(qiáng)于各單項(xiàng)指標(biāo)。第2階段:各指標(biāo)均能監(jiān)測UC療效,Indice的監(jiān)測效能最強(qiáng)。結(jié)論循環(huán)micro RNA能穩(wěn)定高效的診斷UC并監(jiān)測其治療后效果。
[Abstract]:Objective to explore and establish a method of diagnosing ulcerative colitis (ulcerative colitis,UC) based on circulating micro RNA and to monitor its effect after treatment. Methods in the first stage, dextran sulfate (Dextran sulfate sodium,DSS) was used to induce rat UC model. Real-time quantitative RT-PCR screening for differential Serum micro RNA.ELISA Detection of Serum Peripheral Anti-neutrophil Cytoplasmic Antibody (periunclear antineutrophil cytoplasmic antibodies,p ANCA) .Logistic regression to establish a Diagnostic Model Indice. The diagnostic efficacy of (receiver operating characteristic,ROC curve was evaluated. Stage 2: to verify the diagnostic efficacy of UC and evaluate the efficacy of UC. Results in the first stage, the rat UC model was established successfully. Mi R-20bmmi R-21 and let-7e* were highly expressed in the serum of UC rats. There was no significant change in the expression of Mi R-125b-1 *. P ANCA was overexpressed in the serum of UC rats. The diagnostic efficacy of Indice=0 (2.645 脳 mi R-20b) (1.622 脳 mi R21) (0.979 脳 let-7e*) (0.082 脳 p ANCA) was better than that of each single index. Stage 2: each index can monitor the curative effect of UC and Indice is the most effective. Conclusion circulating micro RNA can diagnose UC stably and efficiently and monitor its effect after treatment.
