【摘要】：目的研究解整合素-金屬蛋白酶9(ADAM9)在對(duì)乙酰氨基酚誘導(dǎo)的小鼠急性肝損傷過(guò)程中的表達(dá)變化規(guī)律。方法 50只小鼠隨機(jī)分為2組,正常組10只,實(shí)驗(yàn)組40只。實(shí)驗(yàn)組小鼠腹腔注射對(duì)乙酰氨基酚溶液550 mg·kg-1,6,24,42和72 h后分別摘除小鼠眼球采血、分離血清,同時(shí)分離正常組小鼠血清,檢測(cè)谷草轉(zhuǎn)氨酶和谷丙轉(zhuǎn)氨酶活性;用蛋白印跡和反轉(zhuǎn)錄聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)等方法檢測(cè)正常組小鼠和注射對(duì)乙酰氨基酚后不同時(shí)間點(diǎn)小鼠肝中ADAM9在蛋白和核酸水平的表達(dá)變化。結(jié)果對(duì)乙酰氨基酚注射小鼠后6 h,ADAM9的蛋白和核酸表達(dá)量顯著下降(P0.05),隨著時(shí)間的延長(zhǎng),ADAM9的蛋白和核酸表達(dá)量繼續(xù)下降,24 h達(dá)到最低值(P0.05),然后隨著肝損傷后的修復(fù),ADAM9的表達(dá)量逐漸增加,在注射對(duì)乙酰氨基酚42 h后,ADAM9的蛋白和核酸表達(dá)量增加顯著(P0.05),72 h恢復(fù)到接近正常水平。結(jié)論 ADAM9在對(duì)乙酰氨基酚誘導(dǎo)的小鼠急性肝損傷過(guò)程中表達(dá)呈顯著變化,ADAM9可能起到促進(jìn)肝損傷后修復(fù)的重要作用。
[Abstract]:Objective to study the expression of integrin-metalloproteinase 9 (ADAM9) in paracetamol induced acute liver injury in mice. Methods 50 mice were randomly divided into two groups: normal group (n = 10) and experimental group (n = 40). The mice in the experimental group were injected with paracetamol solution for 550 mg kg-1,6,24,42 and 72 h, respectively. The blood samples were collected from the eyeballs of the mice and the serum was isolated from the normal mice. The activities of glutamic oxaloacetic transaminase and alanine aminotransferase were measured. The expression of ADAM9 in the liver of normal and paracetamol treated mice was detected by Western blot and reverse transcription polymerase chain reaction (RT-PCR) at different time points. Results the protein and nucleic acid expression of ADAM9 decreased significantly 6 h after injection of acetaminophen (P0.05), and the protein and nucleic acid expression of ADAM9 continued to decrease to the lowest level at 24 h (P0.05), and then ADAM9 was repaired with liver injury. The amount of expression gradually increased, The expression of ADAM9 protein and nucleic acid increased significantly (P0.05) at 72 h after injection of paracetamol for 42 h. Conclusion the expression of ADAM9 in acute liver injury induced by acetaminophen may play an important role in promoting the repair of liver injury induced by acetaminophen in mice.
