【摘要】：目的探討枯草桿菌二聯(lián)活菌腸溶膠囊(美常安)對(duì)潰瘍性結(jié)腸炎(UC)大鼠結(jié)腸促炎癥性細(xì)胞因子白細(xì)胞介素(IL)-1β和細(xì)胞黏附分子CD44表達(dá)的影響及其作用機(jī)制。方法采用2,4,6-三硝基苯磺酸(TNBS)/乙醇法灌腸建立UC大鼠模型。將SD大鼠隨機(jī)分成正常對(duì)照組、模型組、美常安治療組。觀察各組疾病活動(dòng)指數(shù)(DAI)、結(jié)腸組織病理學(xué)評(píng)分(HPS)。造模及美常安干預(yù)7 d后處死大鼠,取結(jié)腸行HE染色,觀察鏡下結(jié)腸病理變化。RT-PCR法檢測(cè)各組大鼠腸黏膜細(xì)胞因子IL-1βmRNA表達(dá),流式細(xì)胞儀檢測(cè)各組大鼠結(jié)腸組織中黏附分子CD44的含量。結(jié)果模型組較正常對(duì)照組大鼠結(jié)腸黏膜可見膿性分泌物、糜爛及潰瘍,美常安干預(yù)后顯著改善UC大鼠結(jié)腸黏膜的病理變化,DAI和HPS評(píng)分下降顯著,并明顯降低UC大鼠IL-1βmRNA和CD44的表達(dá)(P0.05)。結(jié)論枯草桿菌二聯(lián)活菌可能通過下調(diào)UC大鼠IL-1βmRNA和CD44的表達(dá)而發(fā)揮干預(yù)作用。
[Abstract]:Objective to investigate the effect of Bacillus subtilis enteric-coated capsule (Meichang an) on the expression of interleukin-1 尾 (IL) 尾) and cell adhesion molecule (CD44) in colonic inflammatory cytokines (IL) 尾) and adhesion molecules (CD44) in rats with ulcerative colitis (UC). SD rats were randomly divided into normal control group, model group and mechangan treatment group. The disease activity index (DAI),) and colonic histopathology score (HPS).) were observed in each group. After 7 days of intervention, the rats were killed, the colons were stained with HE, and the expression of intestinal mucosal cytokine IL-1 尾 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). The content of adhesion molecule CD44 in colonic tissue of rats in each group was detected by flow cytometry. Results the mucosal purulent secretion, erosion and ulceration were observed in the model group compared with the normal control group. The pathological changes of colonic mucosa in UC rats were significantly improved after the intervention of Mechangan. The scores of Dai and HPS decreased significantly in the model group. The expression of IL-1 尾 mRNA and CD44 in UC rats was significantly decreased (P 0.05). Conclusion Bacillus subtilis combined with Bacillus subtilis may play an intervention role by down-regulating the expression of IL-1 尾 mRNA and CD44 in UC rats.
