脂肪間充質(zhì)干細胞經(jīng)肝動脈移植治療晚期肝病

【摘要】:背景:干細胞移植是晚期肝病比較有前景的治療方法,脂肪間充質(zhì)干細胞是繼骨髓間充質(zhì)干細胞后的又一研究熱點。目的:探討經(jīng)肝動脈移植大鼠脂肪間充質(zhì)干細胞對晚期肝病的影響。方法:將45只SD大鼠隨機分為3組,對照組、模型組和移植組各15只。模型組和移植組皮下注射四氯化碳誘導(dǎo)建立大鼠肝硬化模型,移植組大鼠經(jīng)肝動脈注入1 mL CFSE標(biāo)記的脂肪間充質(zhì)干細胞,模型組經(jīng)肝動脈注入等量的生理鹽水,對照組不予處理。移植治療后4周,觀察各組大鼠的肝臟病理組織學(xué)變化,肝功能及肝纖維化程度。結(jié)果與結(jié)論:1在大鼠的肝臟內(nèi)見到綠色熒光標(biāo)記的脂肪間充質(zhì)干細胞。2經(jīng)蘇木精-伊紅染色和Masson染色后發(fā)現(xiàn):模型組大鼠肝小葉結(jié)構(gòu)不清,有假小葉形成,細胞濁腫疏松,部分變性壞死,見炎癥細胞浸潤;對照組大鼠肝組織未見異常。移植組大鼠的肝臟病理組織學(xué)改變介于模型組和對照組之間。3模型組大鼠的血清白蛋白水平低于對照組(P0.05),膽紅素、轉(zhuǎn)氨酶和Ⅳ型膠原水平明顯高于對照組(P0.05);移植組大鼠的血清白蛋白水平高于模型組(P0.05),膽紅素、轉(zhuǎn)氨酶和Ⅳ型膠原低于模型組(P0.05)。4由此可見脂肪間充質(zhì)干細胞移植治療可以改善肝硬化大鼠的肝臟功能,減輕肝纖維化程度。
[Abstract]:Background: stem cell transplantation is a promising treatment for advanced liver disease. Adipose mesenchymal stem cells are another research hotspot after bone marrow mesenchymal stem cells. Objective: to investigate the effect of transhepatic artery transplantation of adipose mesenchymal stem cells on advanced liver disease in rats. Methods: 45 SD rats were randomly divided into 3 groups: control group, model group and transplantation group. The liver cirrhosis model was induced by subcutaneous injection of carbon tetrachloride into the model group and the transplantation group. Rats in the transplantation group were injected with 1 mL CFSE labeled adipose mesenchymal stem cells through the hepatic artery, and the model group were injected with the same amount of normal saline through the hepatic artery. The control group was not treated. Liver histopathology, liver function and liver fibrosis were observed 4 weeks after transplantation. Results and conclusion the green fluorescent labeled adipose mesenchymal stem cells were found in the liver of rats by hematoxylin eosin staining and Masson staining. The results showed that the hepatic lobules in the model group were not clear, pseudolobules were formed, and the cells were turbid and loose. Partial degeneration and necrosis, inflammatory cell infiltration, and no abnormal liver tissue were found in the control group. The liver histopathological changes of the transplantation group were between the model group and the control group. 3.
