S-腺苷蛋氨酸對奈乙硫氰酸酯致大鼠藥物型膽汁淤積性肝病肝纖維化的保護(hù)作用

【摘要】:目的研究S-腺苷蛋氨酸(SAMe)對奈乙硫氰酸酯(ANIT)誘發(fā)大鼠藥物型膽汁淤積性肝病肝纖維化的保護(hù)作用。方法將18只SD大鼠分為3組:A組(空白對照組)、B組(造模+NS)、C組(造模+SAMe),每組各6只,各組大鼠腹腔注射后72h處理。采用全自動生化分析儀檢測各組肝臟血清學(xué)指標(biāo),并取肝臟組織行HE染色及MASSON染色觀察肝臟病理組織學(xué)變化及膠原含量改變。結(jié)果 C組大鼠的血清學(xué)指標(biāo)谷氨酸氨基轉(zhuǎn)移酶(ALT)、天冬氨酸氨基轉(zhuǎn)移酶(AST)、總膽紅素(TBiL)、直接膽紅素(DBiL)、間接膽紅素(IBiL)、谷胺酰轉(zhuǎn)肽酶(γ-GGT)、堿性磷酸酶(ALP)、總膽汁酸(TBA)明顯低于B組,3組之間比較差異均有統(tǒng)計學(xué)意義(P0.05);但3組之間總膽固醇(Chol)水平差異無統(tǒng)計學(xué)意義(P0.05)。C組大鼠的膠原纖維含量及膠原纖維面積比均明顯低于B組,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論 SAMe能通過改善藥物型膽汁淤積性肝病的肝功能,降低膠原纖維含量及面積,進(jìn)一步保護(hù)肝臟組織。
[Abstract]:Objective to study the protective effect of S-adenosylmethionine (SAMe) on hepatic fibrosis induced by neethylthiocyanate (ANIT) in rats with drug-induced cholestatic liver disease. Methods Eighteen Sprague-Dawley rats were divided into 3 groups: group A (blank control group), group B (model NS) and group C (model SAMe), 6 rats in each group. The rats in each group were treated 72 hours after intraperitoneal injection. The liver serological indexes were detected by automatic biochemical analyzer, and liver tissues were stained by HE staining and MASson staining to observe the changes of liver histopathology and collagen content. Results the serum levels of glutamic acid aminotransferase (alt), aspartate aminotransferase (AST), total bilirubin (TBiL), direct bilirubin (DBiL), indirect bilirubin (IBiL), glutamyl transpeptidase (緯 -GGT), alkaline phosphatase (ALP), total bile acid (TBA) were significantly lower in group C rats. There were significant differences between the three groups (P0.05), but there was no significant difference in the level of total cholesterol (Chol) between the three groups (P0.05). The content of collagen fibers and the area ratio of collagen fibers in group C were significantly lower than those in group B (P0.05). The difference was statistically significant (P0.05). Conclusion SAMe can further protect liver tissue by improving liver function, reducing the content and area of collagen fibers in patients with drug-induced cholestatic liver disease.
