功能性胃腸病糞便及腸黏膜菌群與ZO-1蛋白的相關(guān)性研究

【摘要】:目的:探討功能性胃腸病(functional gastrointestinal disorders,FGIDs)患者糞便菌群和腸黏膜菌群的差異性以及二者與腸粘膜中緊密連接蛋白ZO-1的相關(guān)性。方法:通過(guò)結(jié)腸鏡檢查收集32例功能性胃腸病患者的降結(jié)腸黏膜組織以及腸道清潔前糞便標(biāo)本,采用高通量測(cè)序及實(shí)時(shí)熒光定量PCR技術(shù)檢測(cè)糞便中以及腸黏膜中擬桿菌門(Bacteroidetes)、厚壁菌門(Firmicutes)、變形菌門(Proteobacteria)、疣微菌門(Verrucomicrobia)、放線菌門(Actinobacteria)、藍(lán)藻門(Cyanobacteria)、Candidate、梭桿菌門(Fusobacteria)8種菌所占比例,糞便中及腸黏膜定植的擬桿菌門、厚壁菌門、乳酸菌、腸球菌、腸桿菌5種菌的DNA含量與結(jié)腸黏膜組織中緊密連接蛋白ZO-1的m RNA的表達(dá)情況,比較糞便菌群與腸黏膜菌群的差異性,并分析糞便及腸黏膜優(yōu)勢(shì)菌群與緊密連接蛋白ZO-1表達(dá)的相關(guān)性。結(jié)果:腸道糞便菌群與腸黏膜菌群不同,糞便菌群較腸粘膜菌群多樣性(Shannon指數(shù))增加,腸道糞便組為=2.901±0.54,腸粘膜組為=2.29±1.24,P=0.01200.05,有統(tǒng)計(jì)學(xué)意義。在門水平上,糞便菌中以擬桿菌門及厚壁菌門數(shù)量最多,占主要成分;而在腸黏膜組織中變形菌門數(shù)量最多。腸黏膜擬桿菌含量與ZO-1mRNA呈正相關(guān)(r=0.378;P0.05);而糞便擬桿菌與黏膜組織ZO-1mRNA表達(dá)呈負(fù)相關(guān)(r=-0.42,P0.05);組織中乳酸菌和腸桿菌均與ZO-1呈正相關(guān)(r=0.53,P0.01;r=0.355,P0.05)。結(jié)論:腸道菌群包括糞便菌群和粘膜菌群,糞便菌群與粘膜菌群存在差異性,且糞便和黏膜的同一菌群與ZO-1的相關(guān)性不同甚至相悖。
[Abstract]:Objective: to investigate the difference between fecal flora and intestinal mucosal flora in patients with functional gastrointestinal disordersFGIDsand the correlation between them and ZO-1 in intestinal mucosa. Methods: the specimens of descending colon mucosa and feces before intestinal cleaning were collected from 32 patients with functional gastrointestinal disease by colonoscopy. High throughput sequencing and real-time fluorescent quantitative PCR were used to detect eight species of Bacteroides, Firmicuteschus, Proteobacteria, Vertinobacteria, Actinobacteria, Cyanobacteria, and Fusobacteriae in feces and intestinal mucosa. DNA content in fecal and intestinal mucosal colonization of Bacteroides phylum, Mycobacterium thuringiensis, Lactobacillus, Enterococcus and Enterobacter spp., and the expression of tight junction protein ZO-1 mRNA in colonic mucosa. The differences between fecal microflora and intestinal mucosal flora were compared, and the correlation between fecal and intestinal mucosal dominant flora and the expression of tight junction protein ZO-1 was analyzed. Results: the faecal microflora was different from that of intestinal mucosal flora, and the diversity of fecal microflora was higher than that of intestinal mucosal microflora (Shannon index). The intestinal fecal microflora was 2.901 鹵0.54, and the intestinal mucosal group was 2.29 鹵1.24Pu 0.01200.05. there was a significant difference between the fecal microflora and the intestinal mucosal group (P < 0.05). At the portal level, the number of faecal bacteria was the largest in Bacteroides sp. And the phylum of Thinacea was the most, and the number of Proteus in intestinal mucosa was the most. There was a positive correlation between ZO-1 mRNA and ZO-1 mRNA, but a negative correlation between faecal bacillus and ZO-1 mRNA expression, and a positive correlation between Lactic acid bacteria and Enterobacter spp. And ZO-1, 0.355P0.05and 0.373P0.01P0.05P 0.05. the results showed that there was a positive correlation between ZO-1mRNA and ZO-1mRNA, but there was a negative correlation between the expression of ZO-1mRNA and the expression of ZO-1mRNA, and the positive correlation between Lactobacillus and Enterobacter spp. Conclusion: the intestinal flora includes faecal flora and mucosal flora. There are differences between fecal flora and mucosal flora, and the correlation between the same faecal and mucosal flora and ZO-1 is different or even contrary.
