【摘要】：目的探討β-半乳糖苷酶在急性胰腺炎模型大鼠血清中的活性變化水平及意義。方法雄性Wistar大鼠30只,體重200~250 g,隨機分為3組。分別為空白對照組(CON組,n=10)、輕癥急性胰腺炎組(MAP組,n=10)和重癥急性胰腺炎模型組(SAP組,n=10)。分別予以開腹后僅翻動胰十二指腸關腹、主胰管內注射1%�；悄懰徕c溶液(0.1 m L/100 g體重)、主胰管內注射5%牛磺膽酸鈉溶液(0.1 m L/100 g體重)。全自動生化儀檢測各組血清淀粉酶、脂肪酶、丙氨酸氨基轉移酶及鈣離子水平。取胰腺組織4%多聚甲醛固定后切片,HE染色后光鏡下觀察3組胰腺組織病理學改變并評分,免疫組織化學法檢測各組腫瘤壞死因子-α(TNF-α)表達情況。酶聯(lián)免疫吸附(ELISA)法檢測各組血清中β-半乳糖苷酶活性、血清白細胞介素-6水平及胰腺組織β-半乳糖苷酶活性水平。結果與CON組相比較,MAP組和SAP組大鼠血清淀粉酶、脂肪酶、丙氨酸氨基轉移酶、白細胞介素-6水平及胰腺組織β-半乳糖苷酶活性明顯升高(P0.05),血清鈣離子及血清β-半乳糖苷酶活性水平明顯降低(P0.05)。光鏡下可見腺葉組織壞死嚴重、毛細血管擴張充血、組織間隙增寬、大量炎性細胞浸潤等,胰腺組織病理評分明顯升高(P0.05)、TNF-α表達明顯增加。與MAP組相比較,SAP組大鼠血清淀粉酶、脂肪酶、丙氨酸氨基轉移酶、白細胞介素-6水平及胰腺組織β-半乳糖苷酶活性明顯升高(P0.05),血清鈣離子及血清β-半乳糖苷酶活性水平明顯降低(P0.05),胰腺組織壞死嚴重、病理評分升高、TNF-α表達增加。結論血清β-半乳糖苷酶活性水平在大鼠急性胰腺炎時明顯下降,且下降程度與病情嚴重程度有關。
[Abstract]:Objective to investigate the level and significance of 尾-galactosidase in serum of acute pancreatitis rats. Methods Thirty male Wistar rats, weighing 200,250g, were randomly divided into 3 groups. The results were as follows: control group (Con group), mild acute pancreatitis group (map group) and severe acute pancreatitis model group (SAP group). After laparotomy, only pancreaticoduodenal closure was turned over. 1% sodium taurocholate solution was injected into the main pancreatic duct with 0.1 mL / 100 g body weight, and 5% sodium taurocholate solution was injected into the main pancreatic duct with 0.1 mL / 100 g body weight. The levels of serum amylase, lipase, alanine aminotransferase and calcium ion were detected by automatic biochemical instrument. The histopathological changes of pancreas were observed under light microscope after 4% paraformaldehyde fixation and the expression of TNF- 偽 was detected by immunohistochemical method. Serum 尾 -galactosidase activity, serum interleukin-6 level and pancreatic tissue 尾 -galactosidase activity were detected by Elisa. Results compared with CON group, serum amylase, lipase and alanine aminotransferase in map group and SAP group were compared. The level of interleukin-6 and the activity of 尾 -galactosidase in pancreatic tissue increased significantly (P 0.05), and the levels of serum calcium and 尾 -galactosidase decreased significantly (P 0.05). Under the light microscope, there were serious necrosis of glandular lobes, hyperemia of capillaries, widening of tissue space and infiltration of inflammatory cells. The expression of TNF- 偽 in pancreatic tissue was increased significantly. Compared with MAP group, serum amylase, lipase, alanine aminotransferase in SAP group, The level of interleukin-6 and the activity of 尾 -galactosidase in pancreatic tissue were significantly increased (P 0.05), the levels of serum calcium and 尾 -galactosidase were significantly decreased, the pancreatic tissue necrosis was serious, and the expression of TNF- 偽 was increased. Conclusion the level of serum 尾-galactosidase decreased significantly in acute pancreatitis in rats, and the degree of decline was related to the severity of the disease.
