【摘要】:目的:探討TNF-α、IL-6的表達(dá)水平在實(shí)驗(yàn)性急性末端回腸炎中的變化。 方法:選取30只健康成年的雄性雜種犬,運(yùn)用隨機(jī)數(shù)字抽樣法分成6組:正常對(duì)照組(A組)、高溫高濕高血糖組(B組)、正常血糖顱腦火器傷組(C組)、高血糖顱腦火器傷組(D組)、正常血糖濕熱下顱腦火器傷組(E組)、高血糖濕熱下顱腦火器傷組(F組),每組5只。將后四組(C、D、E、F組)給予手槍彈顱腦火器傷造模。在實(shí)驗(yàn)組(B、C、D、E、F組)共五組造模后,將犬立刻仰臥位固定在手術(shù)臺(tái)上,并行氣管插管、心電監(jiān)護(hù)、心肺復(fù)蘇,觀測(cè)各組犬生命體征的變化。觀察過(guò)程中,除心臟擠壓及使用球囊呼吸器搶救外,不做其他處理。在無(wú)菌條件下,立即開(kāi)腹,收集距回盲部1-3cm處的末端回腸組織。肉眼大體觀察,石蠟包埋后行蘇木精-伊紅染色法(HE染色)制作病理切片,并在光學(xué)顯微鏡下觀察其變化,采用逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)檢測(cè)造模后0.5h、1h、3h的TNF-αmRNA、IL-6mRNA表達(dá)水平的變化。 結(jié)果: 1、各組實(shí)驗(yàn)犬肛溫隨時(shí)間的變化實(shí)驗(yàn)組(B、C、D、E、F組)犬的肛溫在各時(shí)間點(diǎn)(10min、1h、2h、3h)均有升高,隨著時(shí)間的推移,肛溫逐漸升高,以F組對(duì)犬的肛溫影響最大,實(shí)驗(yàn)組分別與正常組比較,差異有統(tǒng)計(jì)學(xué)意義(PO.05)。通過(guò)實(shí)驗(yàn),濕熱、高血糖、顱腦火器傷三重因素實(shí)驗(yàn)條件下的肛溫變化較其他兩兩組合的雙重實(shí)驗(yàn)條件更為劇烈,兩兩組合的雙重實(shí)驗(yàn)條件較單純因素的實(shí)驗(yàn)條件下肛溫的變化劇烈。 2、末端回腸組織肉眼及鏡下炎性評(píng)分肉眼觀,實(shí)驗(yàn)組(B、C、D、E、F組)均以急性炎性反應(yīng)為表現(xiàn),且F組較其它組炎性反應(yīng)嚴(yán)重;病理切片鏡下示,實(shí)驗(yàn)組炎性細(xì)胞浸潤(rùn)增多,淋巴管、乳糜管擴(kuò)張,絨毛變粗變短,表現(xiàn)為急性炎性反應(yīng),實(shí)驗(yàn)組分別與正常組比較,差異有統(tǒng)計(jì)學(xué)意義(PO.05)。各實(shí)驗(yàn)組出現(xiàn)炎性評(píng)分隨時(shí)間的延長(zhǎng),其炎性損傷的程度逐漸加重。 3、末端回腸組織TNF-αmRNA的表達(dá)水平各實(shí)驗(yàn)組(B、C、D、E、F組)傷后0.5h、1h、3h,TNF-αmRNA在末端回腸組織的表達(dá)逐漸升高,以F組升高最大,實(shí)驗(yàn)組分別與正常組比較,差異有統(tǒng)計(jì)學(xué)意義(PO.05)。通過(guò)實(shí)驗(yàn)表明,TNF-α可能參與了實(shí)驗(yàn)性急性末端回腸炎的發(fā)病過(guò)程,與時(shí)間呈依賴性,隨著末端回腸組織炎性損傷的加重,其表達(dá)量增加,具有明顯促炎作用。 4、末端回腸組織IL-6mRNA的表達(dá)水平傷后0.5h、1h、3h,實(shí)驗(yàn)組(B、C、D、E、F組)IL-6mRNA在末端回腸組織的表達(dá)依次升高,且隨著時(shí)間延長(zhǎng),升高數(shù)值越大,以F組最明顯,實(shí)驗(yàn)組分別與正常組比較,,差異有統(tǒng)計(jì)學(xué)意義(PO.05)。通過(guò)實(shí)驗(yàn)說(shuō)明,IL-6在實(shí)驗(yàn)性急性末端回腸炎的發(fā)病過(guò)程中與時(shí)間呈依賴性,但其整體表達(dá)水平均較TNF-αmRNA的少,IL-6可能在實(shí)驗(yàn)性急性末端回腸炎的發(fā)病過(guò)程中起著輔助促炎效應(yīng)。 結(jié)論: 1、在實(shí)驗(yàn)性ATI中,隨著時(shí)間的延長(zhǎng),末端回腸組織炎性損傷逐漸加重; 2、TNF-α、IL-6在實(shí)驗(yàn)性ATI中的表達(dá)呈時(shí)間依賴性; 3、TNF-α、IL-6協(xié)同參與了實(shí)驗(yàn)性ATI的發(fā)病過(guò)程。
[Abstract]:Objective : To investigate the expression level of TNF - 偽 and IL - 6 in experimental acute terminal ileitis .
Methods : Thirty healthy adult male hybrid dogs were randomly divided into six groups : normal control group ( group A ) , high - temperature high - humidity hyperglycemia group ( group B ) , normal blood sugar craniocerebral gunshot wound group ( group C ) , high blood glucose and craniocerebral gunshot wound group ( group D ) , normal blood glucose wet - heat and craniocerebral gunshot wound group ( group F ) .
Results :
1 . The anal temperature of dogs in experimental group ( B , C , D , E , F ) increased with time ( 10min , 1h , 2h , 3h ) .
2 . At the end ileum , the visual and microscopic inflammatory scores were observed . The experimental groups ( group B , C , D , E , F ) were characterized by acute inflammatory reaction , and the F group had more inflammatory response than other groups .
Under the pathological section , the inflammatory cell infiltration in experimental group increased , the lymphatic vessels , the breast erosions were dilated and the villi became shorter and shorter . The experimental group showed acute inflammatory reaction . The experimental group had significant difference compared with the normal group ( PO.05 ) . The inflammatory score of each experimental group increased with time , and the degree of inflammatory injury was gradually increased .
3 . The expression level of TNF - 偽 mRNA in the terminal ileum was 0.5 h , 1 h , 3 h , and TNF - 偽 mRNA increased gradually in the terminal ileum . The results showed that TNF - 偽 might be involved in the pathogenesis of experimental acute terminal ileitis .
4 . IL - 6 mRNA expression in terminal ileum was 0.5h , 1h , 3h . The expression of IL - 6 mRNA in the terminal ileum was increased in the experimental group . The results showed that IL - 6 had a statistically significant difference with time in the pathogenesis of experimental acute terminal ileitis , but the expression level of IL - 6 was lower than that of normal group . IL - 6 could play an auxiliary pro - inflammatory effect in the pathogenesis of experimental acute terminal ileitis .
Conclusion :
1 . In the experimental ATI , with the extension of time , the inflammatory injury of terminal ileum was gradually increased ;
2 . The expression of TNF - 偽 and IL - 6 in experimental ATI was time - dependent ;
