TNF-a、IL-6在實驗性急性末端回腸炎中的表達

【摘要】:目的:探討TNF-α、IL-6的表達水平在實驗性急性末端回腸炎中的變化。 方法:選取30只健康成年的雄性雜種犬,運用隨機數(shù)字抽樣法分成6組:正常對照組(A組)、高溫高濕高血糖組(B組)、正常血糖顱腦火器傷組(C組)、高血糖顱腦火器傷組(D組)、正常血糖濕熱下顱腦火器傷組(E組)、高血糖濕熱下顱腦火器傷組(F組),每組5只。將后四組(C、D、E、F組)給予手槍彈顱腦火器傷造模。在實驗組(B、C、D、E、F組)共五組造模后,將犬立刻仰臥位固定在手術(shù)臺上,并行氣管插管、心電監(jiān)護、心肺復(fù)蘇,觀測各組犬生命體征的變化。觀察過程中,除心臟擠壓及使用球囊呼吸器搶救外,不做其他處理。在無菌條件下,立即開腹,收集距回盲部1-3cm處的末端回腸組織。肉眼大體觀察,石蠟包埋后行蘇木精-伊紅染色法(HE染色)制作病理切片,并在光學(xué)顯微鏡下觀察其變化,采用逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)檢測造模后0.5h、1h、3h的TNF-αmRNA、IL-6mRNA表達水平的變化。 結(jié)果: 1、各組實驗犬肛溫隨時間的變化實驗組(B、C、D、E、F組)犬的肛溫在各時間點(10min、1h、2h、3h)均有升高,隨著時間的推移,肛溫逐漸升高,以F組對犬的肛溫影響最大,實驗組分別與正常組比較,差異有統(tǒng)計學(xué)意義(PO.05)。通過實驗,濕熱、高血糖、顱腦火器傷三重因素實驗條件下的肛溫變化較其他兩兩組合的雙重實驗條件更為劇烈,兩兩組合的雙重實驗條件較單純因素的實驗條件下肛溫的變化劇烈。 2、末端回腸組織肉眼及鏡下炎性評分肉眼觀,實驗組(B、C、D、E、F組)均以急性炎性反應(yīng)為表現(xiàn),且F組較其它組炎性反應(yīng)嚴重;病理切片鏡下示,實驗組炎性細胞浸潤增多,淋巴管、乳糜管擴張,絨毛變粗變短,表現(xiàn)為急性炎性反應(yīng),實驗組分別與正常組比較,差異有統(tǒng)計學(xué)意義(PO.05)。各實驗組出現(xiàn)炎性評分隨時間的延長,其炎性損傷的程度逐漸加重。 3、末端回腸組織TNF-αmRNA的表達水平各實驗組(B、C、D、E、F組)傷后0.5h、1h、3h,TNF-αmRNA在末端回腸組織的表達逐漸升高,以F組升高最大,實驗組分別與正常組比較,差異有統(tǒng)計學(xué)意義(PO.05)。通過實驗表明,TNF-α可能參與了實驗性急性末端回腸炎的發(fā)病過程,與時間呈依賴性,隨著末端回腸組織炎性損傷的加重,其表達量增加,具有明顯促炎作用。 4、末端回腸組織IL-6mRNA的表達水平傷后0.5h、1h、3h,實驗組(B、C、D、E、F組)IL-6mRNA在末端回腸組織的表達依次升高,且隨著時間延長,升高數(shù)值越大,以F組最明顯,實驗組分別與正常組比較,,差異有統(tǒng)計學(xué)意義(PO.05)。通過實驗說明,IL-6在實驗性急性末端回腸炎的發(fā)病過程中與時間呈依賴性,但其整體表達水平均較TNF-αmRNA的少,IL-6可能在實驗性急性末端回腸炎的發(fā)病過程中起著輔助促炎效應(yīng)。 結(jié)論: 1、在實驗性ATI中,隨著時間的延長,末端回腸組織炎性損傷逐漸加重; 2、TNF-α、IL-6在實驗性ATI中的表達呈時間依賴性; 3、TNF-α、IL-6協(xié)同參與了實驗性ATI的發(fā)病過程。
[Abstract]:Objective : To investigate the expression level of TNF - 偽 and IL - 6 in experimental acute terminal ileitis .

Methods : Thirty healthy adult male hybrid dogs were randomly divided into six groups : normal control group ( group A ) , high - temperature high - humidity hyperglycemia group ( group B ) , normal blood sugar craniocerebral gunshot wound group ( group C ) , high blood glucose and craniocerebral gunshot wound group ( group D ) , normal blood glucose wet - heat and craniocerebral gunshot wound group ( group F ) .

Results :

1 . The anal temperature of dogs in experimental group ( B , C , D , E , F ) increased with time ( 10min , 1h , 2h , 3h ) .

2 . At the end ileum , the visual and microscopic inflammatory scores were observed . The experimental groups ( group B , C , D , E , F ) were characterized by acute inflammatory reaction , and the F group had more inflammatory response than other groups .
Under the pathological section , the inflammatory cell infiltration in experimental group increased , the lymphatic vessels , the breast erosions were dilated and the villi became shorter and shorter . The experimental group showed acute inflammatory reaction . The experimental group had significant difference compared with the normal group ( PO.05 ) . The inflammatory score of each experimental group increased with time , and the degree of inflammatory injury was gradually increased .

3 . The expression level of TNF - 偽 mRNA in the terminal ileum was 0.5 h , 1 h , 3 h , and TNF - 偽 mRNA increased gradually in the terminal ileum . The results showed that TNF - 偽 might be involved in the pathogenesis of experimental acute terminal ileitis .

4 . IL - 6 mRNA expression in terminal ileum was 0.5h , 1h , 3h . The expression of IL - 6 mRNA in the terminal ileum was increased in the experimental group . The results showed that IL - 6 had a statistically significant difference with time in the pathogenesis of experimental acute terminal ileitis , but the expression level of IL - 6 was lower than that of normal group . IL - 6 could play an auxiliary pro - inflammatory effect in the pathogenesis of experimental acute terminal ileitis .

Conclusion :

1 . In the experimental ATI , with the extension of time , the inflammatory injury of terminal ileum was gradually increased ;


2 . The expression of TNF - 偽 and IL - 6 in experimental ATI was time - dependent ;
