TNFSF14及其受體LTβR和HVEM在病毒肝炎中的作用

【摘要】:目的:探討TNFSF14(即LIGHT)及其受體LTβR和HVEM在暴發(fā)性病毒肝炎中的作用。方法:MHV-3感染易感小鼠建立暴發(fā)性病毒肝炎模型,通過(guò)HE染色和血清ALT水平的測(cè)定比較了LIGHT KO和WT兩組小鼠的肝損情況;定量PCR技術(shù)檢測(cè)了MHV-3感染C57BL/6小鼠后各時(shí)相點(diǎn)(0 h、12 h、24 h、48 h、72 h)肝臟和脾臟組織HVEM和LTβR的mRNA水平。流式細(xì)胞術(shù)檢測(cè)了臨床病毒性重癥肝炎病人PBMC中HVEM和LTβR的表達(dá)情況。結(jié)果:MHV-3感染72 h后,WT小鼠的肝臟出現(xiàn)明顯壞死灶,而LIGHT KO小鼠的肝臟則未見(jiàn)異常;LIGHT KO小鼠血清中的ALT濃度也顯著低于WT組(P0.01)。MHV-3感染48 h后,C57BL/6小鼠脾臟中HVEM及LTβR的mRNA水平均有顯著升高(P0.05);肝臟中LTβR表達(dá)在12 h時(shí)就有顯著上調(diào)(P0.01)。與之一致的是,臨床重癥肝炎病人PBMC中HVEM和LTβR的表達(dá)較正常人均有明顯升高。結(jié)論:TNFSF14可能通過(guò)與其受體LTβR和HVEM的相互作用在病毒誘發(fā)的暴發(fā)性肝炎中扮演重要角色。
[Abstract]:Aim: to investigate the role of TNFSF14 and its receptors LT 尾 R and HVEM in fulminant viral hepatitis. Methods the fulminant viral hepatitis model was established in susceptible mice infected with MHV-3. The liver damage of LIGHT KO and WT groups were compared by HE staining and serum ALT level. The mRNA levels of HVEM and LT 尾 R in liver and spleen tissues of C57BL/6 mice infected with MHV-3 were determined by quantitative PCR. The expression of HVEM and LT 尾 R in PBMC of patients with severe viral hepatitis was detected by flow cytometry. Results 72 hours after infection, liver necrosis was observed in the mice with WMHV-3 infection. However, the serum ALT concentration of LIGHT KO mice was significantly lower than that of WT group P0.01U. MHV-3. The mRNA levels of HVEM and LT 尾 R in spleen of C57BL / 6 mice were significantly higher than those of WT group 48 h after infection, and the expression of LT 尾 R increased significantly at 12 h. Similarly, the expression of HVEM and LT 尾 R in PBMC of patients with severe hepatitis was significantly higher than that in normal controls. Conclusion: TNFSF14 may play an important role in viral induced fulminant hepatitis by interacting with its receptor LT 尾 R and HVEM.
