幽門螺桿菌感染小鼠胃黏膜組織細胞因子變化的實驗研究

【摘要】:目的研究幽門螺旋桿菌(簡稱幽門螺桿菌)感染小鼠胃黏膜組織細胞因子變化,初步探討幽門螺桿菌感染的免疫機制。方法選取8~10周齡BALB/c小鼠24只,雌雄各半,分為觀察組和對照組,兩組造模前均禁食12h,觀察組以2×109CFU/mL濃度的幽門螺桿菌菌液0.5mL/d灌胃,連續(xù)7d。對照組灌胃給生理鹽水,0.5mL/d,連續(xù)7d,進行幽門螺桿菌感染小鼠胃黏膜造模。采用實時定量熒光PCR(Real-time PCR)檢測檢測感染2、4周后,小鼠血漿中γ干擾素(IFN-γ)、重組改構(gòu)人腫瘤壞死因子α(TNF-α)、白細胞介素-8(IL-8)以及IL-10mRNA的表達量,并于感染4周后,處死小鼠,無菌取各組小鼠的胃組織,采用酶聯(lián)免疫吸附試驗夾心法檢測胃黏膜組織中IFN-γ、TNF-α、IL-8及IL-10蛋白的水平。結(jié)果兩組小鼠感染2周后,觀察組IL-8高于對照組,差異有統(tǒng)計學意義(P0.05)。感染4周后,IFN-γ、TNF-α、IL-8的mRNA及蛋白表達量均明顯上升,但觀察組上升幅度明顯高于對照組,差異有統(tǒng)計學意義(P0.05)。結(jié)論TH2細胞在幽門螺桿菌感染中的作用表現(xiàn)不明顯,能促進TH1型細胞細胞因子的表達,引發(fā)TH1為主的免疫反應(yīng)。
[Abstract]:Objective to study the changes of cytokines in gastric mucosa of mice infected with Helicobacter pylori (Helicobacter pylori) and to explore the immune mechanism of Helicobacter pylori infection. Methods 24 10-week-old BALB/c mice, half male and half female, were divided into two groups: the observation group and the control group. The two groups fasted for 12 hours before modeling. The observation group was fed with 2 脳 109CFU/mL Helicobacter pylori solution 0.5mL/d for 7 days. The control group was given normal saline 0.5 mL / d for 7 days to model the gastric mucosa of mice infected with Helicobacter pylori. The expression of IFN- 緯, recombinant human tumor necrosis factor- 偽 (TNF- 偽), interleukin-8 (IL-8) and IL-10mRNA in plasma of mice were detected by real-time fluorescence PCR(Real-time 4 weeks after infection. The mice were killed after 4 weeks of infection. The gastric tissues of each group were collected and the levels of IFN- 緯 TNF- 偽 TNF- 偽 IL-8 and IL-10 protein in gastric mucosa were detected by enzyme-linked immunosorbent assay (Elisa). Results after 2 weeks of infection, the IL-8 in the observation group was higher than that in the control group, and the difference was statistically significant (P 0.05). After 4 weeks of infection, the expression of mRNA and protein of TNF- 偽 TNF- 偽 IL-8 in the observation group was significantly higher than that in the control group, and the difference was statistically significant (P 0.05). Conclusion the effect of TH2 cells on Helicobacter pylori infection is not obvious, which can promote the expression of TH1 cytokines and induce the immune response of TH1.
