自噬在實(shí)驗(yàn)性急性胰腺炎中變化的研究

【摘要】:目的: 探討自噬在實(shí)驗(yàn)性急性胰腺炎中的變化,通過(guò)觀察自噬在急性胰腺炎發(fā)病過(guò)程的變化情況,說(shuō)明自噬與急性胰腺炎的關(guān)系,進(jìn)一步為急性胰腺炎的發(fā)病機(jī)制提供新的理論。 方法: 將SD大鼠54只,按照隨機(jī)分配原則將大鼠分為三組,對(duì)照組(SO),急性水腫性胰腺炎(AEP),急性壞死性胰腺炎(ANP),采用胰膽管逆行注射牛磺膽酸鈉方法制造模型。每組大鼠按不同的時(shí)相點(diǎn)分別于術(shù)后第3、6、12h麻醉后心臟采血,留取血清檢測(cè)淀粉酶;并將胰腺組織小部分做電鏡檢測(cè);小部分用4%甲醛固定,常規(guī)切片和HE染色;采用免疫組化方法檢測(cè)LC3B、P62表達(dá)。 結(jié)果: 1、胰腺病理學(xué)評(píng)分: ①AEP組、ANP組和SO組之間各時(shí)間點(diǎn)病理?yè)p傷評(píng)分差異有顯著性(P0.05);②AEP組病理?yè)p傷評(píng)分開(kāi)始增加,6h與12hAEP組無(wú)顯著差異(P0.05);③ANP組損傷評(píng)分明顯增加,隨時(shí)間增加損傷程度增加,各時(shí)間點(diǎn)差異有顯著性(P0.05)。 2、血清淀粉酶變化: ①SO組各時(shí)間點(diǎn)之間血淀粉酶值無(wú)顯著差異(P0.05);②AEP組、ANP組較SO組值有升高,ANP組中較AEP組升高,且差異有顯著性(P0.05)。 3、胰腺組織電鏡下表現(xiàn): 我們觀察3h各組電鏡下的表現(xiàn):3hAEP組和3hANP組較SO組自噬體和自噬溶酶體數(shù)量明顯增加(P0.05),,3hANP組較3hAEP組自噬體和自噬溶酶體數(shù)量明顯增加(P0.05)。 4、胰腺組織LC3B免疫組化染色評(píng)分: ①SO組可見(jiàn)LC3B少量表達(dá),且各時(shí)間點(diǎn)無(wú)顯著差異(P0.05);AEP組、ANP組LC3B表達(dá)增強(qiáng),與SO組比較差異有顯著性(P0.05);②AEP組中LC3B表達(dá)在6h和12h表達(dá)開(kāi)始明顯增加,與3h比較差異有顯著性(P0.05)。③ANP組LC3B表達(dá)明顯增加,染色加深,且隨著時(shí)間的延長(zhǎng),表達(dá)逐漸增加,與AEP組同一時(shí)間點(diǎn)比較差異有顯著性(P0.05)。 5、胰腺組織P62免疫組化染色評(píng)分: ①SO組見(jiàn)少量P62表達(dá),且各時(shí)間點(diǎn)無(wú)明顯差異(P0.05);AEP組、ANP組P62表達(dá)增強(qiáng),與SO組比較有差異顯著性(P0.05);②但同一時(shí)間點(diǎn)AEP組與ANP組間P62表達(dá)無(wú)顯著差異性,AEP組和ANP組中各時(shí)間點(diǎn)之間無(wú)顯著差異(P0.05)。 結(jié)論: 1、經(jīng)胰膽管逆行注射1%、5%;悄懰徕c成功復(fù)制了AEP和ANP動(dòng)物模型。 2、AEP組和ANP組中自噬的表達(dá)是增加的,且ANP組中自噬的表達(dá)較AEP組中表達(dá)增加,ANP組和AEP組中自噬的表達(dá)隨病理?yè)p傷加重而增加。 3、AEP組和ANP組中自噬的降解水平減弱。 4、自噬的功能損傷可能在急性胰腺炎的發(fā)病過(guò)程中起著一定作用。
[Abstract]:Objective: To explore the changes of autophagy in experimental acute pancreatitis, and to explain the relationship between autophagy and acute pancreatitis by observing the changes of autophagy in the pathogenesis of acute pancreatitis, and to provide a new theory for the pathogenesis of acute pancreatitis. Methods: Fifty-four SD rats were divided into three groups according to the principle of random distribution: control group, acute edematous pancreatitis (AEPN) and acute necrotizing pancreatitis (ANPN). The model was established by retrograde injection of sodium taurocholate into the pancreatic duct. In each group, the blood samples were collected from the heart at different time points at 6h after anesthesia, and the serum amylase was collected, and a small portion of pancreatic tissue was examined by electron microscopy, and a small portion was fixed with 4% formaldehyde, and stained with routine sections and HE. Immunohistochemical method was used to detect the expression of LC _ 3B ~ (2 +) P _ (62). Results: 1. Pancreatic pathological score: There was significant difference in pathological injury score between 1AEP group and so group at different time points. There was no significant difference in pathological injury score between 1AEP group and 12hAEP group at 6 h. There was significant difference between different time points (P 0.05). 2, the changes of serum amylase: There was no significant difference in serum amylase value between different time points in 1SO group. The serum amylase level in AEP group was higher than that in so group. The level of amylase in ANP group was higher than that in AEP group, and the difference was significant (P 0.05). 3. The histologic features of the pancreas were as follows: At 3 h, the number of autophagy and lysosome in AEP group and 3hANP group were significantly increased than those in so group. The number of autophagy and lysosome in 3hAEP group was significantly higher than that in 3hAEP group. 4. LC3B immunohistochemical staining score of pancreatic tissue: A small amount of LC3B expression was observed in the 1SO group, and there was no significant difference at each time point. The LC3B expression in the 1SO group increased significantly compared with that in the so group, and the expression of LC3B began to increase at 6 and 12 hours. Compared with 3h, the expression of LC3B in P0.05An 路3ANP group was obviously increased, the staining was deepened, and the expression increased gradually with the prolongation of time, and there was a significant difference between P0.05ANP group and AEP group at the same time point. 5, P62 immunohistochemical staining score of pancreatic tissue: A small amount of P62 expression was found in 1SO group, and there was no significant difference at each time point. There was no significant difference in the expression of P62 between AEP group and ANP group at the same time point. There was no significant difference between AEP group and ANP group at different time points. Conclusion: 1. The animal models of AEP and ANP were successfully established by retrograde injection of 5% sodium taurocholate through the pancreaticobiliary duct. 2the expression of autophagy was increased in AEP group and ANP group, and the expression of autophagy in ANP group was higher than that in AEP group. 3 the degradation level of autophagy in AEP group and ANP group was decreased. 4, the function of autophagy may play a role in the pathogenesis of acute pancreatitis.
