藍(lán)莓對(duì)小鼠急性酒精性脂肪肝TNF-α-R1和SREBP-1c影響的初步研究

【摘要】:目的:觀察藍(lán)莓對(duì)急性酒精性脂肪肝(alcoholic fatty liver,AFL)小鼠肝組織中腫瘤壞死因子-α-受體1(tumor necrosis factor-α-recepter1,TNF-α-R1)、膽固醇調(diào)節(jié)元件結(jié)合蛋白(sterol regulatory element-binding protein,SREBP)-1c m RNA及蛋白表達(dá)的影響。方法:50只C57BL/6雄性小鼠隨機(jī)分為空白組、模型組、藍(lán)莓汁高劑量組(20.0 g/kg)、藍(lán)莓汁低劑量組(10.0 g/kg)、硫普羅寧組(30 mg/kg),每組10只。除空白組外,其余各組小鼠分別予Lieber-De Carli酒精飼料喂飼,藍(lán)莓汁高、低劑量組及硫普羅寧組同時(shí)分別予相應(yīng)劑量藍(lán)莓汁或硫普羅寧灌胃1次/d,連續(xù)15 d,第16天給予1次乙醇灌胃,建立急性AFL動(dòng)物模型。全自動(dòng)生化分析儀測(cè)定血清丙氨酸氨基轉(zhuǎn)移酶(alanine transaminase,ALT)、天冬氨酸氨基轉(zhuǎn)移酶(aspartate transaminase,AST)、甘油三酯(triglycerides,TG)及膽固醇(cholesterol,TC),酶聯(lián)免疫吸附法檢測(cè)血清TNF-α,光鏡下觀察小鼠肝臟病理學(xué)變化,RTPCR、Western blot檢測(cè)小鼠肝組織中TNF-α-R1、SREBP-1c m RNA及蛋白質(zhì)表達(dá)。結(jié)果:藍(lán)莓汁高、低劑量組和硫普羅寧組小鼠血清ALT分別為(67.70±5.86)、(81.66±4.93)、(63.49±3.27)U/L,AST分別為(184.46±14.57)、(214.45±22.03)、(169.16±18.52)U/L,TG分別為(0.88±0.06)、(1.22±0.08)、(0.81±0.09)mmol/L,TC分別為(2.30±0.13)、(2.74±0.10)、(2.21±0.16)mmol/L,TNF分別為(49.14±6.14)、(66.55±5.67)、(44.68±5.39)ng/L,均顯著低于模型組(P=0.000或P=0.025),肝細(xì)胞病理學(xué)改變明顯輕于模型組(P=0.000);藍(lán)莓汁高、低劑量組和硫普羅寧組小鼠肝組織TNF-α-R1 m RNA表達(dá)分別為100.44±9.51、132.70±8.07、94.02±6.05,蛋白表達(dá)分別為0.52±0.04、0.72±0.04、0.49±0.04,SREBP-1c m RNA表達(dá)分別為133.52±5.91、174.51±10.62、129.83±9.83,蛋白表達(dá)分別為0.26±0.05、0.41±0.03、0.23±0.05,均明顯低于模型組(均P=0.000)。結(jié)論:藍(lán)莓能改善小鼠急性AFL,可能與下調(diào)其肝組織TNF-α-R1與SREBP-1c的表達(dá)有關(guān)。
[Abstract]:Objective: to observe the effect of blueberry on the expression of TNF- 偽 -R1G, regulatory element-binding protein SSREBPN-1c m RNA and protein in liver of mice with acute alcoholic fatty liver (fatty livera). Methods: TNF- 偽 -receptor 1(tumor necrosis factor- 偽 -receptor 1 (TNF- 偽 -R1) and cholesterol regulatory element binding protein (regulatory element-binding) were detected in mice liver tissues. Methods Fifty male C57BL/6 mice were randomly divided into blank group, model group, high dose group (20.0 g / kg), low dose group (10.0 g / kg), tiopronin group (30 mg / kg), each group (10 rats). In addition to the blank group, the other mice were fed with Lieber-De Carli alcohol feed, the blueberry juice was high, the low-dose group and the tiopronin group were given the corresponding dose of blueberry juice or tiopronin by stomach for one once a day for 15 days, and on the 16th day, the rats were given ethanol. Acute AFL animal model was established. Serum alanine transaminase (Alanine transaminase), aspartate transaminase (aspartate transaminase), triglyceridesy (TGG) and cholesterol (TC) were measured by automatic biochemical analyzer. Serum TNF- 偽 was detected by enzyme-linked immunosorbent assay (Elisa) and pathological changes of mouse liver were observed under light microscope.
