色氨酸緩解甲氨蝶呤引起的大鼠腸道損傷

【摘要】:本試驗(yàn)評(píng)價(jià)了色氨酸對(duì)由甲氨蝶呤(MTX)引起大鼠損傷的腸道形態(tài)結(jié)構(gòu)、抗氧化能力和機(jī)體代謝的影響。通過(guò)灌胃不同劑量色氨酸(Trp),分析大鼠血生化指標(biāo)、氧化應(yīng)激相關(guān)指標(biāo)、血紅素氧合酶-1和內(nèi)源性代謝物等的變化,探討色氨酸對(duì)腸道損傷的保護(hù)作用。 方法:選用SD雄性大鼠,體重為200~220g。大鼠預(yù)飼一周后按體重隨機(jī)分為6組,每組8只:正常對(duì)照組(NC組)、色氨酸對(duì)照組(TRP組)、甲氨蝶呤模型組(MTX組)、低劑量色氨酸+甲氨蝶呤組(L-TRP+MTX組)、中劑量色氨酸+甲氨蝶呤組(M-TRP+MTX組)、高劑量色氨酸+甲氨蝶呤組(H-TRP+MTX組)。預(yù)飼一周后,不同劑量色氨酸干預(yù)組大鼠分別灌胃等體積的100、200和300mg/kg.bw的色氨酸;正常對(duì)照組和色氨酸對(duì)照組分別灌喂等體積的蒸餾水和200mg/kg.bw的色氨酸,每日按劑量灌胃一次,連續(xù)7天;第4天靜脈注射單劑量甲氨蝶呤(10mg/kg.bw),連續(xù)兩天,,對(duì)照組注射等體積的生理鹽水。 實(shí)驗(yàn)結(jié)果:(1)色氨酸能改善腸道形態(tài)結(jié)構(gòu),其中低、中劑量色氨酸的效果更顯著(P0.05);(2)色氨酸使血漿中D-乳酸含量降低(P0.05);(3)氧化應(yīng)激導(dǎo)致大鼠腸組織氧化還原狀態(tài)失衡,使MDA和MPO水平顯著增加(P0.01);色氨酸有助于提高其抗氧化能力,低、中劑量色氨酸對(duì)于提高抗氧化能力效果更顯著(P0.05);(4)通過(guò)色氨酸干預(yù)能顯著提高HO-1的表達(dá),其中低、中劑量色氨酸干預(yù)效果更好(P0.05);(5)氧化應(yīng)激導(dǎo)致機(jī)體代謝紊亂,色氨酸使大鼠血清中葡萄糖和脯氨酸含量降低,琥珀酸和檸檬酸含量升高(P0.05)。 結(jié)論:色氨酸能改善腸道形態(tài)結(jié)構(gòu),提高組織抗氧化能力,調(diào)節(jié)機(jī)體代謝。色氨酸能有效地保護(hù)組織器官免受氧化損傷,這可能與HO-1的表達(dá)有關(guān)。
[Abstract]:The effects of tryptophan on the intestinal morphology, antioxidant capacity and metabolism of rats induced by methotrexate (MTX) were evaluated. The changes in blood biochemical indexes, oxidative stress related indexes, heme oxygenase -1 and endogenous metabolites were analyzed by different doses of tryptophan (Trp), and tryptophan was used to investigate the effect of tryptophan on intestine. Protective effect of canal injury.
Methods: SD male rats were divided into 6 groups according to weight of 200 ~ 220g. rats, each group was divided into 6 groups according to weight, 8 rats in each group: normal control group (group NC), tryptophan control group (group TRP), methotrexate group (MTX group), low dose tryptophan + methotrexate group (L-TRP+ MTX group), middle dose tryptophan + methotrexate group (M-TRP+MTX group), high dose Tryptophan + methotrexate group (group H-TRP+MTX). After a week of prefeeding, 100200 and 300mg/kg.bw tryptophan of different doses of tryptophan intervention group were given respectively. The normal control group and tryptophan control group were fed with equal volume of distilled water and tryptophan 200mg/kg.bw, respectively, once daily for 7 days at a dose, and fourth days. Intravenous injection of single dose of methotrexate (10mg/kg.bw) for two consecutive days, the control group was injected with equal volume of normal saline.
The results were as follows: (1) tryptophan could improve the morphological structure of intestinal tract, of which low dose tryptophan was more effective (P0.05); (2) tryptophan decreased the content of D- lactate (P0.05) in plasma (P0.05); (3) oxidative stress caused the imbalance of redox state of intestinal tissue in rats and increased the level of MDA and MPO significantly (P0.01); tryptophan helped to improve its oxygen resistance. The effect of tryptophan is more significant (P0.05) in improving the antioxidant capacity (P0.05); (4) the intervention of tryptophan can significantly improve the expression of HO-1, which is lower, the middle dose of tryptophan is better (P0.05); (5) oxidative stress causes metabolic disorder in the body, and tryptophan reduces the content of glucose and proline in the serum of rats, succinic The content of perperic acid and citric acid increased (P0.05).
Conclusion: tryptophan can improve the morphological structure of the intestine, improve the antioxidant capacity of the tissue and regulate the metabolism of the body. Tryptophan can protect the tissues and organs from oxidative damage effectively, which may be related to the expression of HO-1.
