【摘要】：目的:觀察大葉茜草素(mollugin)對大鼠肝星狀細(xì)胞系CFSC-2G活化和膠原合成的影響并探討其分子機(jī)制。方法:小劑量(10μmol/L)過氧化氫(H_2O_2)誘導(dǎo)CFSC-2G細(xì)胞30 min后,再加入不同濃度(0、20、40、60和120μmol/L)的mollugin處理。MTT法檢測細(xì)胞活力,real-time PCR和Western blot法分別檢測核因子E2相關(guān)因子2(Nrf2)、血紅素加氧酶-1(HO-1)、核因子κB(NF-κB)p65、Bcl-2、Bcl-x L、Bax以及肝星狀細(xì)胞活化標(biāo)志物α-平滑肌肌動(dòng)蛋白和Ⅰ型膠原蛋白的mRNA和蛋白表達(dá),并用Western blot法檢測p38絲裂原活化蛋白激酶(p38MAPK)的磷酸化水平。結(jié)果:低劑量H_2O_2可以誘導(dǎo)CFSC-2G細(xì)胞活化,mollugin明顯促進(jìn)p38 MAPK磷酸化,上調(diào)Nrf2和HO-1的mRNA和蛋白表達(dá),下調(diào)NF-κB p65、Bcl-2和Bcl-x L的mRNA和蛋白表達(dá),抑制H_2O_2誘導(dǎo)活化的CFSC-2G細(xì)胞活力和膠原合成(P0.05)。結(jié)論:Mollugin可能通過上調(diào)Nrf2和HO-1并下調(diào)NF-κB p65和Bcl-2表達(dá),抑制CFSC-2G細(xì)胞活化和膠原合成。
[Abstract]:Aim: to observe the effects of Rubidium grandiflorum on the activation of CFSC-2G and collagen synthesis in rat hepatic stellate cell line and to explore its molecular mechanism.Methods: low dose of 10 渭 mol / L H _ 2O _ 2 (H _ 2O _ 2) was used to induce CFSC-2G cells for 30 min.MRNA and protein expression of actin and type I collagen,The phosphorylation level of p38 mitogen activated protein kinase (p38 MAPK) was detected by Western blot method.Results: low dose H_2O_2 could induce the activation of CFSC-2G cells to promote the phosphorylation of p38 MAPK, up-regulate the expression of mRNA and protein of Nrf2 and HO-1, down-regulate the expression of mRNA and protein of NF- 魏 B p65- Bcl 2 and Bcl-x L, and inhibit the activation of CFSC-2G cells induced by H_2O_2 and the synthesis of collagen.Conclusion: by up-regulating the expression of Nrf2 and HO-1 and down-regulating the expression of NF- 魏 B p65 and Bcl-2, the activation and collagen synthesis of CFSC-2G cells may be inhibited by w: Mollugin.
