急性肝衰竭小鼠回腸、結腸以及糞便菌群變化的研究

【摘要】:目的:探討急性肝衰竭小鼠腸道微生態(tài)菌群的變化,尋找特征性差異菌,為急性肝衰竭腸道微生態(tài)紊亂的診治提供依據(jù)。方法:BALB/c雄鼠30只(空白組5只,模型組25只),D-氨基半乳糖制造小鼠急性肝衰竭模型。提取小鼠下消化道不同節(jié)段(回腸、結腸)內(nèi)容物及糞便的微生物DNA,進行PCR擴增,對16S V3-V4區(qū)進行高通量測序,利用生物信息學分析技術對測序結果進行OTU聚類,物種注釋,α多樣性分析,LEf Se(LDA Effect Size)分析,尋找模型組特征性差異菌。結果:2組小鼠,共存活10只,其中模型組死亡率為80%。回腸、結腸內(nèi)容物及糞便共30個樣本,測序覆蓋深度97%。α多樣性分析顯示急性肝衰竭小鼠回腸中細菌豐度增加(chao1指數(shù)105.7±18.4與149.3±55,P0.01),多樣性增加(shannon指數(shù)2.5±0.1與2.8±0.8,P0.05);結腸中細菌豐度減低(chao1指數(shù)285.4±46.1與276.98±46.7,P0.05)及多樣性增加(shannon指數(shù)4.9±0.3與5.65±0.39,P0.01),糞便中細菌多樣性降低(shannon指數(shù)5.1±0.3與4.87±0.29,P0.01),豐度(chao1指數(shù)270.1±53.7與250.2±54.4,P=0.067)無顯著差異。模型組腸道菌群與空白組相比小鼠腸道中檢測到10個菌門,以厚壁菌門、擬桿菌門和變形菌門為主。模型組回腸中細菌門水平差異無統(tǒng)計學意義;結腸中擬桿菌門比例增加(9.3%±7.9%與33.8%±18.2%,P=0.0240.05),TM7菌門比例增加(0.2%±0.3%與2.2%±1.6%,P=0.0290.05);糞便中厚壁菌門減少(32.8%±20.5%與10.2%±5.5%,P=0.05)、擬桿菌門增加(57.3%±23.4%與86.1%±67.2%,P=0.07),但差異無統(tǒng)計學意義。在最佳分類水平上,回腸梭菌科豐度減少(44.95%±19.28%與7.51%±16.57%,P0.05),結腸理研菌科(1.08%±1.01%與4.18%±2.39%,P0.05)、S24-7(4.75%±4.87%與22.77%±13.05%,P0.05)、F16(0.24%±0.28%與2.18%±1.61%,P0.05)等豐度增加。LEf Se(LDA Effect Size)分析顯示兩組在葡萄球菌科、S24-7方面差異有統(tǒng)計學意義,其中S24-7可定義為特征性差異菌群。結論:不同節(jié)段腸道擁有不同的腸道菌群結構,急性肝衰竭小鼠腸道微生態(tài)紊亂,回腸菌群過度生長。急性肝衰竭與S24-7過度生長有相關性。
[Abstract]:Objective: to investigate the changes of intestinal microecological flora in mice with acute liver failure and to find out the characteristic differential bacteria to provide evidence for the diagnosis and treatment of intestinal microecological disorder in acute liver failure.Methods the acute liver failure model was established in 30 male mice (5 in the blank group and 25 in the model group) with D-galactosamine (D-galactosamine).The microbial DNA of different segments of the lower digestive tract (ileum, colon) and feces of mice were extracted and amplified by PCR. The 16s V3-V4 region was sequenced by high-throughput sequencing. The sequencing results were clustered by OTU using bioinformatics analysis technique.Species annotation, 偽 -diversity analysis and LEF Se(LDA Effect size analysis were used to find the characteristic differential bacteria in the model group.Results there were 10 mice survived in the 2 groups, and the mortality of the model group was 80%.There were 30 samples of ileum, colon contents and feces.There was no significant difference in bacterial diversity between 4.9 鹵0.3 and 5.65 鹵0.39 (P 0.01), but there was no significant difference in bacterial diversity in feces between 5.1 鹵0.3 and 4.87 鹵0.29 (P 0.01) and 270.1 鹵53.7 and 250.2 鹵54.4 (P 0.067).Ten phylum were detected in the intestinal tract of the model group compared with that in the blank group, and the main ones were phylum thunbergiformis, Bacteroides sp., and Proteus phylum.妯″瀷緇勫洖鑲犱腑緇嗚弻闂ㄦ按騫沖樊寮傛棤緇熻瀛︽剰涔,
