【摘要】：目的分析非酒精性脂肪肝患者(NAFLD組)與正常對照組(HC組)唾液菌群的多樣性。方法收集24例脂肪肝患者和22例正常志愿者的唾液標(biāo)本,提取總DNA,用PCR擴(kuò)增16S rDNAV4-V5區(qū),并進(jìn)行高通量測序,將測序數(shù)據(jù)進(jìn)行OTU聚類、多樣性分析及物種注釋等。結(jié)果兩組納入對象的唾液菌群獲得的序列平均有39 118條,其中NAFLD組聚類OTUs約308±48,HC組聚類OTUs約305±53。多樣性分析NAFLD組和HC組示整體唾液菌群的多樣性方面差異較小。物種分析數(shù)據(jù)表明唾液菌群在門及屬水平的組成基本相同,但相對豐度不同,NAFLD組在門水平示厚壁菌門(20.42%)豐度比例升高,擬桿菌門(36.75%)比例降低;屬水平的梭菌屬、卟啉單胞菌屬、韋永氏球菌屬的比例較高,而普氏菌屬、普雷沃菌屬比例顯著低。結(jié)論 NAFLD組相較于HC組的唾液中厚壁菌門的菌群豐度相對升高而擬桿菌門的菌群豐度相對減少,推測唾液菌群失衡與NAFLD發(fā)病可能存在一定關(guān)系。
[Abstract]:Objective to analyze the diversity of salivary flora in patients with nonalcoholic fatty liver (NAFLD) and control group (HC). Methods the total DNA was extracted from 24 patients with fatty liver and 22 healthy volunteers. The 16s rDNAV4-V5 region was amplified by PCR. High throughput sequencing was carried out. OTU clustering, diversity analysis and species annotation were performed on the sequencing data. Results the average number of sequences obtained from the two groups of salivary flora was 39,118. Among them, the OTUs of NAFLD group was about 308 鹵48? HC group? OTUs was about 305 鹵53. The diversity analysis of NAFLD group and HC group showed little difference in the diversity of the whole saliva flora. The species analysis data showed that the composition of salivary flora was basically the same at the phylum and genus level. But in NAFLD group with different relative abundance, the proportion of abundance increased and the proportion of Bacteroides 36.75) decreased at the gate level, while the proportion of Clostridium, porphyromonas, Wayonella was higher, while that of Pueraria was higher. Conclusion compared with HC group, the abundance of sputum of NAFLD group is higher than that of HC group, and the abundance of Bacteroides phylum is relatively lower. It is suggested that the imbalance of salivary flora may be related to the pathogenesis of NAFLD.
