【摘要】：目的:前期研究表明酪酸梭菌對損傷的胃黏膜具有保護(hù)作用,但具體效應(yīng)物質(zhì)不明。本文通過檢測酪酸梭菌的主要代謝產(chǎn)物氫氣和丁酸對損傷胃黏膜的效應(yīng),探究黏膜保護(hù)作用的具體機制。方法:將雄性ICR小鼠通過乙醇灌胃誘導(dǎo)胃黏膜損傷,并以氫氣和丁酸分別進(jìn)行處理,觀測胃組織的大體變化和組織結(jié)構(gòu)損傷情況,通過RT-q PCR檢測炎癥因子白細(xì)胞介素12(IL-12)、Ras相關(guān)核蛋白1(RAN1)和單核細(xì)胞趨化蛋白1(MCP-1)的含量,采用免疫組化法觀測Bcl-2和Bax蛋白表達(dá)量變化。結(jié)果:胃組織大體觀測發(fā)現(xiàn)丁酸能夠保護(hù)胃黏膜,而氫氣不具有此作用;HE染色也提示丁酸能夠顯著改善乙醇造成的胃黏膜病理損傷。RT-q PCR則顯示所檢測炎癥因子IL-12、RAN1和MCP-1的表達(dá)量較模型對照組有顯著降低(P0.01)。在丁酸組,Bax的表達(dá)量較模型組顯著減低(P0.01),而Bcl-2的含量卻明顯提高(P0.01)。結(jié)論:酪酸梭菌胃黏膜保護(hù)效應(yīng)的主要產(chǎn)物可能是丁酸而不是氫氣。丁酸能夠通過抑制炎癥反應(yīng),下調(diào)Bax/Bcl-2表達(dá)比例,從而實現(xiàn)對胃黏膜的保護(hù)作用。
[Abstract]:Objective: previous studies have shown that Clostridium caseinate has protective effect on gastric mucosal injury, but the specific substance is unknown. This paper studied the effects of hydrogen and butyric acid, the main metabolites of Clostridium caseinate, on gastric mucosal injury. Methods: gastric mucosal injury was induced by ethanol in male ICR mice and treated with hydrogen and butyric acid respectively. RT-q PCR was used to detect the levels of Ras associated nucleoprotein 1 (RAN1) and monocyte chemoattractant protein 1 (MCP-1). The changes of Bcl-2 and Bax protein expression were observed by immunohistochemical method. Results: the gastric mucosa was protected by butyric acid. The results of HE staining also suggested that butyric acid could significantly improve the pathological injury of gastric mucosa induced by ethanol. RT-q PCR showed that the expression of IL-12, IL-12, RAN1 and MCP-1 in butyric acid group was significantly lower than that in the model control group (P 0.01). The expression of Bax was significantly lower than that of the model group, while the content of Bcl-2 was significantly increased. Conclusion: butyric acid may be the main product of gastric mucosal protective effect of Clostridium caseinate, but not hydrogen. Butyric acid can inhibit inflammatory reaction. The expression of Bax/Bcl-2 was down-regulated to protect gastric mucosa.
