【摘要】：目的觀察腫瘤壞死因子(TNF)-α對腎血管收縮的影響及機制。方法 56只大鼠制備離體灌注腎模型,隨機分為8組(n=7):A1組:Kreb液灌流;A2組:Kreb液+TNF-α灌流;B1組:Kreb液+Verapamil灌流;B2組:Kreb液+Verapamil+TNF-α灌流;C1組:無鈣Kreb液灌流;C2組:無鈣Kreb液+TNF-α灌流;D1組:無鈣Kreb液+2-APB灌流;D2組:無鈣Kreb液+2-APB+TNF-α灌流。各組均在刺激期加內(nèi)皮素(ET)。灌流結(jié)束后,計算腎臟水腫率,HE染色觀察腎小球及腎小管形態(tài)、結(jié)構(gòu)。結(jié)果各組基礎(chǔ)灌注壓比較無顯著差異(P0.05)。A1、A2、B1、B2、C1、C2組ET刺激后,腎灌注壓較基礎(chǔ)壓均明顯升高(P0.05);A2、B2、C2組灌注壓升高值分別顯著高于A1、B1、C1組(P0.01)。D1、D2組ET刺激后,腎灌注壓均略升高,但與基礎(chǔ)灌注壓比較無顯著差異(P0.05);兩組灌注壓升高值比較無顯著性差異(P0.05)。8組腎臟的水腫率均低于30%,灌流后腎臟標本切片均未發(fā)現(xiàn)明顯的器質(zhì)性損傷,與灌流前相符。結(jié)論 TNF-α可能通過上調(diào)1,4,5-三磷酸肌醇受體(IP3R)增強ET引起的腎血管收縮。
[Abstract]:Objective to observe the effect and mechanism of TNF- 偽 on renal vasoconstriction in 56 rats. Eight groups were randomly divided into 8 groups: 1% Kreb solution, 2% Kreb solution, A 2 group, 1% Kreb solution, TNF- 偽 perfusion group, 1% Kreb solution, Verapamil solution, 2% Kreb solution, 2% Kreb solution, Verapamil, 2% Kreb solution, 1 group, 2 + calcium free Kreb solution, and C2 group, no calcium Kreb solution, TNF- 偽 perfusion group, no calcium Kreb solution, 2 APB perfusion fluid, D 2 group, D 2 group, D 2 group, respectively. Calcium free Kreb solution 2-APB TNF- 偽 was perfused. The morphology and structure of glomeruli and renal tubules were observed by HE staining. Results there was no significant difference in basal perfusion pressure among the three groups. The renal perfusion pressure was significantly higher than that of the base pressure. After et stimulation, the renal perfusion pressure in the P0.05A2OB2OC2 group was significantly higher than that in the A1-B1OC1 group, and the renal perfusion pressure increased slightly after et stimulation. However, there was no significant difference between the two groups compared with the basic perfusion pressure (P 0.05), and there was no significant difference in the increase of perfusion pressure between the two groups. The edema rate of kidney was lower than that of 30% in the P0.05 group, and no significant organic injury was found in the sections of the renal specimens after perfusion. Conclusion TNF- 偽 may enhance the renal vasoconstriction induced by et by upregulating the receptor IP _ 3R.
