人羊膜上皮細(xì)胞移植治療肝纖維化免疫大鼠

【摘要】:目的觀察人羊膜上皮細(xì)胞(human amniotic epithelial cells,h AECs)在正常免疫大鼠體內(nèi)存活、遷徙及對大鼠肝纖維化程度的影響,為h AECs應(yīng)用于臨床提供實(shí)驗(yàn)依據(jù)。方法 10周齡雄性SD大鼠64只,體重220~280 g,隨機(jī)分為4組,每組16只。A組為肝纖維化+細(xì)胞移植(移植細(xì)胞量4×106個(gè))組,B組為肝纖維化+生理鹽水組,C組為肝纖維化組,D組為空白對照組。移植后2周,取A組大鼠心、肝、脾、腎、肺、腦組織采用DNA-PCR法檢測人Alu基因重復(fù)序列,免疫組織化學(xué)染色觀察人白細(xì)胞抗原G(hu man leucocyte antigen G,HLA-G)表達(dá),并比較各臟器陽性表達(dá)百分比(脾臟為細(xì)胞注入器官不納入統(tǒng)計(jì));取各組肝組織,行HE染色觀察并采用肝纖維化組織學(xué)半定量計(jì)分系統(tǒng)(SSS)行肝纖維化半定量分析,行TGF-β1免疫組織化學(xué)染色觀察并行免疫組織化學(xué)評分(immunohistochemical score,ISH);取各組血清行谷草轉(zhuǎn)氨酶(aspartate transaminase,AS T)、谷丙轉(zhuǎn)氨酶(alanine aminotransferase,ALT)、白蛋白(albumin,ALB)濃度檢測,定量分析大鼠肝纖維化程度。結(jié)果A組各臟器中人Alu基因重復(fù)序列及HLA-G均表達(dá)陽性,肝組織中陽性表達(dá)百分比顯著高于其余組織,比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。A、B、C組肝纖維化組織學(xué)半定量計(jì)分分別為(10.47±3.20)、(13.84±3.46)、(13.85±3.16)分,A組顯著低于B、C組(P0.05);B、C組間差異無統(tǒng)計(jì)學(xué)意義(P0.05)。A、B、C、D組TGF-β1的ISH評分分別為(3.60±1.50)、(5.38±2.60)、(5.50±2.40)、(1.87±1.36)分,A、B、C組顯著高于D組,A組顯著低于B、C組(P0.05);B、C組間差異無統(tǒng)計(jì)學(xué)意義(P0.05)。A、B、C組血清AST、ALT濃度顯著高于D組,A組顯著低于B、C組(P0.05),B、C組間差異無統(tǒng)計(jì)學(xué)意義(P0.05);ALB濃度顯著低于D組,A組顯著高于B、C組(P0.05);B、C組間差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 h AECs可在正常免疫大鼠體內(nèi)存活,可緩解肝纖維化程度并改善血清肝功能指標(biāo)。
[Abstract]:Objective To observe the effect of human amniotic epithelial cells (human amniotic epithelial cells, H AECs) on the survival of normal immune rats, migration and the influence on the degree of hepatic fibrosis in rats, H AECs provide an experimental basis for clinical application.
