焦磷酸測序法和Sanger測序法檢測丙型肝炎病毒基因分型的方法學比較

【摘要】:目的探討焦磷酸測序法檢測丙肝分型在臨床應用中的準確性和敏感性,評價其方法的優(yōu)缺點。方法收集100例丙肝患者的血漿標本,分裝相同的2份,通過焦磷酸測序法和Sanger測序法分別進行丙型肝炎病毒基因分型的檢測,對兩種測序方法的結(jié)果進行比較。結(jié)果焦磷酸測序法和Sanger測序法檢測HCVR N A基因分型時,100例標本中,92例結(jié)果一致,8例結(jié)果不一致,一致率為92%。其中,HCV-R N A病毒載量1×104IU/ml有80例,其測序結(jié)果完全一致為100%,HCV-R N A病毒載量1×104 IU/ml有20例,其中焦磷酸測序共檢測20例,Sanger測序法共檢測14例,12例結(jié)果相同。結(jié)論焦磷酸測序法檢測丙型肝炎病毒基因分型,不僅結(jié)果準確可靠,而且與Sanger測序法比較,其敏感性更高。此外,焦磷酸測序還可以進行定量分析。
[Abstract]:Objective to investigate the accuracy and sensitivity of pyrosequencing in the clinical application of hepatitis C typing, and to evaluate the advantages and disadvantages of the method. The genotyping of hepatitis C virus was detected by pyrosequencing and Sanger sequencing. Results the results of two sequencing methods were compared. Results in the detection of HCVR na genotyping by pyrosequencing and Sanger sequencing, 92 cases were consistent with the results of 8 cases, and the consistency rate was 92%. Among them, 80 cases had HCV-R N A viral load of 1 脳 10 4 u / ml. There were 20 cases of HCV-R A virus carrying 1 脳 10 ~ 4 IU/ml, of which 20 cases were detected by pyrosequencing and 12 cases were detected by pyrosequencing. Conclusion pyrosequencing for HCV genotyping. The results are not only accurate and reliable, but also more sensitive than Sanger sequencing. In addition, pyrosequencing can also be used for quantitative analysis.
