【摘要】：目的:分析幽門螺桿菌誘發(fā)十二指腸潰瘍患者血清中B淋巴細(xì)胞活化因子(BAFF)表達(dá)情況,探討B(tài)AFF對(duì)Th17細(xì)胞分化的影響。方法:ELISA法測(cè)患者血清中BAFF與IL-17濃度;MTT法測(cè)T細(xì)胞與幽門螺桿菌共培養(yǎng)后存活率;流式細(xì)胞術(shù)測(cè)BAFF-R表達(dá);RT-PCR、Western blot、ELISA法測(cè)RORγt和RORα及IL-17、IL-21、IL-22的m RNA和蛋白表達(dá)。結(jié)果:與對(duì)照相比,幽門螺桿菌陽性患者血清中BAFF與IL-17表達(dá)水平顯著增高(P0.05),兩者表達(dá)水平呈正相關(guān)(r=0.87,P0.01)。BAFF處理后,幽門螺桿菌誘導(dǎo)的T細(xì)胞生存活力較對(duì)照增強(qiáng),BAFF-R表達(dá)增加,Th17細(xì)胞標(biāo)志性轉(zhuǎn)錄因子RORγt和RORα及細(xì)胞因子IL-17、IL-21、IL-22的m RNA和蛋白表達(dá)水平均明顯高于對(duì)照(P0.05)。結(jié)論:幽門螺桿菌能引起十二指腸潰瘍患者體內(nèi)BAFF高表達(dá),進(jìn)而促進(jìn)Th17細(xì)胞分化。
[Abstract]:Objective: to analyze the expression of B lymphocyte activating factor (BAFF) in serum of patients with duodenal ulcer induced by Helicobacter pylori. To investigate the effect of BAFF on the differentiation of Th17 cells. Methods the survival rate of T cells and Helicobacter pylori co-cultured was measured by BAFF and IL-17 concentration in serum of patients. Flow cytometry was used to detect the expression of ROR 緯 t and ROR 偽, and the m RNA and protein of ROR 偽 and IL-17 IL-21, IL-22. Results: compared with the control group, the expression of BAFF and IL-17 in the serum of patients with Helicobacter pylori positive was significantly higher than that in the control group. The expression of BAFF and IL-17 in serum of patients with Helicobacter pylori positive was significantly higher than that of the control group. The survival activity of T cells induced by Helicobacter pylori was significantly higher than that of the control group. The expression levels of ROR 緯 t and ROR 偽, as well as the expression levels of m RNA and protein of IL-17, IL-21, IL-22 in Th17 cells were significantly higher than those in the control group P0.050.Conclusion: Helicobacter pylori is more potent than the control group. Can cause high expression of BAFF in patients with duodenal ulcer. And then promote the differentiation of Th17 cells.
