先天性小耳畸形微小RNA綜合調(diào)控網(wǎng)絡(luò)的生物信息學(xué)分析

目的構(gòu)建以miRNA為中心的綜合調(diào)控網(wǎng)絡(luò),探討關(guān)鍵miRNA、靶基因、轉(zhuǎn)錄因子、長鏈非編碼RNA與先天性小耳畸形發(fā)病間的關(guān)系,為進一步明確病因提供新的研究思路。方法分析小耳畸形殘耳軟骨與健側(cè)耳軟骨間miRNAs差異表達譜,基于24個差異表達的miRNAs,預(yù)測靶基因、轉(zhuǎn)錄因子和長鏈非編碼RNA,構(gòu)建綜合調(diào)控圖并對關(guān)鍵節(jié)點進行生物信息學(xué)分析。結(jié)果基于24個差異miRNAs,共預(yù)測到154個靶基因,209個轉(zhuǎn)錄因子和22個長鏈非編碼RNA。MiR-221/222-3p、miR-320c、miR-3122、miR-4521,靶基因ZEB2、TRPS1、GNAI3、SHOC2、PDPK1、PDPK1,轉(zhuǎn)錄因子EP300、STAT1、FOS、KMT2A、RNF2和長鏈非編碼RNA TUG1、MIAT間存在相互調(diào)節(jié)作用構(gòu)成關(guān)鍵模塊。MiR-221/222-3p可與靶基因TRPS1、ZEB2,轉(zhuǎn)錄因子FOS、EP300、STAT1及l(fā)ncRNA MIAT相互調(diào)控,靶基因ZEB2與轉(zhuǎn)錄因子STAT1在調(diào)控網(wǎng)絡(luò)中作為關(guān)鍵節(jié)點可連接多個靶點構(gòu)成綜合調(diào)控網(wǎng)絡(luò)。結(jié)論初步建立了先天性小耳畸形綜合調(diào)控網(wǎng)絡(luò),找到了...
